细胞间粘附因子-1(ICAM-1)在自身免疫性内耳病中的作用

摘　　要：目的探讨细胞间粘附因子-1(ICAM-1)在自身免疫性内耳病中的作用。方法选用耳廓反射灵敏的健康豚鼠70只,随机分为三组:Ⅰ组:对照组(10只),不施加任何因素;Ⅱ组:佐剂组(20只),首次皮下注射完全弗氏佐剂(CFA)0.1m l,以后每周皮下注射1次0.1m l不完全弗氏佐剂,共注射6次,分别于末次注射后第3d和2周处死7只;Ⅲ组:实验组(40只),首次皮下注射鸡Ⅱ型胶原(400μg)与CFA的乳浊液0.1m l,以后每周皮下注射Ⅱ型胶原。各组动物均在处死前测定听觉诱发电位(ABR),并作扫描电镜观察和内耳免疫组织化学测ICAM-1的表达情况。结果Ⅲ组ABR各波潜伏期及Ⅰ-Ⅲ波波间期较Ⅰ组和Ⅱ组延长(P<0.05),Ⅲ组3d及2周时第三排外毛细胞部分听毛缺失和倒伏;Ⅰ组和Ⅱ组耳蜗组织ICAM-1表达为阴性;Ⅲ组螺旋神经节细胞、神经纤维、血管纹、基底膜和血管内皮细胞等部位均有ICAM-1表达;Ⅲ组2周较3d时ICAM-1表达明显下降。结论自身免疫性内耳病发病初期炎症因子ICAM-1起重要作用,其可能是此病的发病机制之一。Objective To study the effect of intercellular adhesion molecule （ ICAM - 1 ） on the autoimmune inner ear disease. Methods 70 healthy guinea were randomly divided into 3 groups. GroupⅠ （10 guinea pigs）, the control group,were under no treatment. Group Ⅱ （20 guinea pigs）, the collagenic group, at the first time were injected subcutaneously with 0.1 ml complete Freundadjuvant（CFA） ,and at the following 5 times were injected subcutaneously with 0. 1 ml incomplete Freundadjuvant（IFA） weekly. 7 guinea pigs were sacrificed respectively on 3rd and 14th day after the last injection. Group Ⅲ （40 guinea pigs）,the experimental group, at the first time were injected subcutaneously with 0.1 ml the emulsion of CC Ⅱ （4001xg） and CFA, and at the following 5 times were injected subcutaneously with 0.1 ml the emulsion of CC Ⅱ （200μg） and IFA weekly. Results The latencies of all waves and the interpeak latencies of Ⅰ wave- Ⅲ wave in group Ⅲ had significance compared with those in groupⅠ and group Ⅱ （ P 〈 0.05 ）. In group Ⅲ, the cilia of outer hair cells of the third row lodged or lost on the 3rd day and 14th day. The inner ears in groupⅠ and group Ⅱ almost had no expression of ICAM - 1. There was expression of ICAM - 1 in the spiral ganglion cells, the stria vascularis,the nerve fiber,the basilar membrane and the vascular endothelial cells in group Ⅲ ,and furthermore the expression decreased on 14th day than those on 3rd day. Conclusion Icam - 1 plays an important role in the early stage of AIED, it may be one of the mechanisms of AIED.
