摘  要:目的:采用薄膜分散法合并硫酸铵梯度法制备盐酸川芎嗪脂质体,考察各因素对包封率的影响。方法:用HPLC法测定包封率,考察了磷脂和胆固醇用量比例,脂质和十八胺用量,透析时间,硫酸铵浓度以及载药温度对包封率的影响。结果:脂质体的包封率随磷脂-胆固醇用量比例的提高而提高,最高达54.8%;并随脂质用量增大而增大;适量的十八胺用量以及硫酸铵浓度有利于提高包封率;最合适的栽药温度为37℃。结论:薄膜分散法合并硫酸铵梯度法在适当的条件下可制得包封率较高的盐酸川芎嗪脂质体。Objective: To prepare ligustrazine hydrochloride liposome by the ammonium sulfate gradient technology with thin film method and study the infuence of different faetors on the entrapment efficiency. Method: The entrapment efficiency was determined by HPLC. The effects of various factors on the entrapment efficiencies, including phospholipid-cholesterol ratio, lipid and catadecylamine quantity, the dialyse time, the ammonium sulfate concentration, and the test temperature were investigated. Result: The entrapment efficiency was inhanced by the increased phospholipid-cholesterol ratio( the highest entrapment efficiency of 54.8% ) and lipid quantity. The relevant octadecylamine quantity and ammonium sulfate concentration made for higher entrapment efficiency. The best test temperature was 37℃. Conclusion: The ligustrazine hydrochloride liposome with high entrapment efficiency was prepared by the ammonium sulfate gradient technology with thin film method in relevant condition.
