摘 要:目的 探讨2,5-己二酮(HD)对神经组织神经丝(NF)mRNA水平的影响。方法Wistar雄性大鼠27只,分为3组(1个对照组,2个染毒组),每组9只,经腹腔染毒,剂量分别为200和400mg/(kg·d),连续8周。逆转录聚合酶链反应(RT-PCR)方法检测坐骨神经、脊髓、大脑组织中高分子量NF(NF-H)、中分子量NF(NF-M)和低分子量NF(NF-L)mRNA水平。结果 200和400mg/kg剂量染毒后,坐骨神经NF-L mRNA水平分别为对照组的126%(P〈0.05)和152%(P〈0.01),NF-M mRNA未检测到,NF-HmRNA未发生明显变化;在脊髓、大脑组织中,NF-L mRNA下降为对照组的34%-54%(P〈0.01),NF-M mRNA未发生明显变化,400mg/kg剂量组中NF-H mRNA下降为对照组的85%(P〈0.05)。结论 HD染毒导致大鼠神经组织中NF亚单位mRNA水平的变化。Objective To investigate the alterations in mRNA levels of neurofilament (NF) in rats intoxicated with 2, 5- hexanedione( HD). Methods HD was administered by intraperitoneal injection to wistar rats with daily dosing of 200 and 400 mg/kg for continuous 8w(five times per week), mRNA levels of light molecular NF( NF-L), medium molecular NF(NF-M) and high molecular NF (NF-H) in sciatic nerve, spinal cord, and cerebrum were determined respectively by RT-PCR. Results mRNA levels of NF-L in rat sciatic nerves were increased to 126% (P 〈 O.05)and 152% (P 〈 0.01 ) at the dosage of 200 and 400 mg/kg individually. For the spinal cord and cerebrum, the expression of NF-L mRNA were both decreased obviously to 34% - 54% ( P 〈 O. 1 ) relatively, and NF- H mRNA of 400 mg/kg dosage were declined to 85 % (P 〈 O. 05 ) of the control. The others were kept relatively unchanged. Conclusion HD-intoxication was associated with alterations in mRNA levels of NF.
