摘  要:目的研究大鼠残肾模型肾脏组织学和缓激肽B2受体(BKB2R)mRNA表达水平的变化。方法24只雄性Wistar大鼠随机分成模型组和假手术组,每组12只。5/6肾切除术构建大鼠残肾模型。于造模前和造模后15、30、60、120 d分别检测大鼠尾动脉血压和血清肌酐水平。造模后1月和4月分别处死模型组和假手术组大鼠各6只,肾组织石蜡切片、PAS染色观察肾脏组织学改变,RT-real time PCR法检测肾组织BKB2R mRNA的表达水平。结果模型组大鼠造模后15 d血压和血清肌酐水平即较造模前明显升高(均P<0.01),以后逐渐上升至120 d达高峰;造模后1月残肾已出现肾小球代偿肥大,系膜中度增生,肾组织BKB2R mRNA的表达水平下调,4月时残肾出现明显的肾小球硬化、肾小管坏死和肾间质纤维化,肾组织BKB2R mRNA的表达水平进一步下调(P<0.01)。结论残肾模型大鼠在造模后15d即出现明显的肾功能不全,随后进行性加重,肾组织BKB2R mRNA的表达水平则逐渐下调。Objective To observe the changes of renal histology and bradykinin B2 receptor expression in rats with chronic renal failure induced by surgery. Methods Twenty four male Wistar rats were divided into sham-operation and experiment group randomly (n= 12 in each). The remnant kidney model was established by 2-stage 5/6 nephrectomy. The blood pressure and serum creatine were obtained before and 15, 30, 60, 120 days after operation respectively. Six rats each in sham operation group and experiment group were killed at 1st and 4th month after the second operation respectively. Remnant kidneys were isolated. The fixed renal tissues were embedded in paraffin, and sectioned into 4 μm-thick slices. The slices were stained by PAS. The expression of BKB2 R mRNA in renal tissues was detected by using RT-real time PCR. Results Blood pressure and serum creatine were increased significantly 15 days after the second operation (both P〈0.01), and reached the peak at 120th day. One month after operation, histological changes were found and the expression of BKB2 R mRNA was significantly downregulated. Four months after operation, there existed a significant renal sclerosis and the expression of BKB2 R mRNA was reduced constantly (P〈0.01). Conclusion A significant renal dysfunction could be produced 15 days after 5/6 nephrectomy. There were a proceeding renal sclerosis and down-regulation of BKB2 R mRNA 4 months after 5/6 nephrectomy.
