摘 要:建立了一种新的制备手性2-辛醇的方法——双菌氧化还原耦联转化外消旋2-辛醇制备(R)-2-辛醇。通过筛选得到了一株选择性氧化外消旋2-辛醇中(S)-2-辛醇的菌株CandidacylindraceaATCC14830,其产物的光学纯度e.e.>98%,(R)-2-辛醇的产率>46%。与报道的一株还原2-辛酮得到(R)-2-辛醇的菌株OenococcusoeniCECT4730氧化还原耦联,底物浓度可达40g/L,产物的e.e.>98%,产率>92%。To develop a practical process for (R)-2-octanol production from racemic 2-octanol, a two step microbial reaction was investigated. The microbial reaction used the eantioselective oxidation of (S)-2-octanol by Candida cylindracea ATCC 14830 and the asymmetric reduction of oxidated 2-octanone by Oenococcus oeni CECT 4730. O. oeni CECT 4730 was a strain reported which can reduce 2-octanone to (R)-2-octanol, C. cylindracea ATCC 14830 was screened as an candidate biocatalyst for preparation of (R)-2-octanol and 2-octanone with the optical purity of 98%e.e. and the yield of 46%. After redox-coupling, the yield and optical purity were 92% and 98% respectively. The substrate concentration was 40g/L.
