PRRSV抗GP5和抗M蛋白抗抗体的制备及免疫作用的研究

摘　　要：制备猪繁殖与呼吸综合征病毒(PRRSV)抗抗体(Ab2),探讨Ab2在不同的动物体内诱导机体产生免疫作用的研究。用已纯化的抗PRRSV-GP5(GP5-Ab1)单抗和抗PRRSV-M(M-Ab1)单抗免疫哈尔滨大白兔,当免疫血清琼扩效价达1∶16时,则加强免疫1次心脏采血分离血清,分别纯化兔抗抗PRRSV-GP5 IgG(GP5-Ab2)和抗抗PRRSV-M IgG(M-Ab2)血清。用GP5-Ab2、M-Ab2以及GP5-Ab2与M-Ab2混合型3种分别免疫昆明小鼠和未免PRRS疫苗的小猪,同时设PRRS弱毒苗、灭活苗和空白组作对照。免疫后第21 d分别采集小鼠血清和猪血清,用PRRSV作抗原包被,间接ELISA检测免疫Ab2和PRRS疫苗的小鼠血清和猪血清全为阳性,空白组仍然为阴性。说明制备的Ab2在不同动物体内均能引起机体产生免疫反应,从而为猪繁殖与呼吸综合征病新型疫苗的研究提供了新的思路。This work included prepared PRRSV＇s anti - antibody （ Ab2 ）, discussed the modulating impact to organism immunity of Ab2 in differ animal. Using purified anti PRRSV - GP5 （ GP5 - Ab1 ） and anti PRRSV- M（M- Ab1） protein antibody to immunize Haerbin＇s big white rabbit, detecting the value, when the value achieved more than 1： 16, after another intensified immunity, blooding from heart, separating and purifying the rabbit anti antibody PRRSV -GPSIgG（ GP5 -Ab2） and PRRSV -MIgG（ M -Ab2 ） serum, using GP5 - Ab2, M - Ab2, the mixture of GP5 - Ab2 and M - Ab2 immunized Kunming mice and no - immune with PRRS vaccine＇s piglet, set PRRS weaken - virulence vaccine and set the blank as the controls, the 21th day after immunity, blooding serum, the indirect ELISA detection results showed the kun - ming mice and piglet＇s serum immunity with Ab2 and PRRS vaccine were all positive, the controls were negative. The results showed that Ab2 in vivo had the function of replacing the PRRSV induced the differ organism mmunity , and these take a new way of the organism to avoid PRRSV＇s infection.
