摘  要:目的建立二甲双弧的高效液相色谱法及其片剂的血药浓度测定方法。方法采用高效液相色谱法测定二甲双胍血药浓度。色谱柱为Hypersil ODS(250mm×4.6mm,5μm),流动相为水。乙腈(70:30,内含0.014moL/L磷酸氢二胺,0.003moL/L十二烷基硫酸钠,加冰醋酸调pH值为6.8),流速为1.2mL/min,检测波长232nm。结果二甲双胍血药浓度为50~2000μg/L,浓度与峰面积比线性良好,相关系数r=0.9990,最低检测浓度为50μg/L。高中低浓度样品的日内、日间测定RSD均〈10%,绝对回收率均〉70%。结论本方法简便、灵敏、准确、可靠,可作为二甲双胍血药浓度的测定方法。Objective To establish a HPLC method to measure the plasma concentration of metformin. Methods A Hypersil-ODS column (250 mm×4.6mm,5μm) was selected. Mobile phase was a mixture of water and acetonitrile (70 : 30, v/v,0. 014 moL/L(NH4)2HPO4, 0. 003 moL/L CH3(CH2)11SO4Na,pH=6.8) with flow rate of 1.2 mL/min. The detection wavelength was 232 nm. Results The standard curve was linear over the range of 50-2 000 μg/L of metformin in plasma (r=0.999 0). The limit of detection was 50μg/L. The recovery rate was more than 70% and the RSD within a day and between days did not exceed 10%, respectively. Conclusion The method is simple, sensitive, accurate and reliable which can be used in the pharmaco- kinetics study of metformin.
