摘  要:目的研究骨髓间质干细胞移植于SD大鼠急性心肌梗死后对心肌细胞凋亡的影响。方法SD大鼠开胸结扎前降支中段30min后,实验组在心肌缺血区域注射Dill标记的骨髓间质干细胞3×100个,对照组以100μL生理盐水作心肌缺血区域注射。4~6周后取心脏组织切片,采用TUNEL法和免疫组织化学技术进行心肌细胞凋亡的原位检测,以及肌钙蛋白T(Troponin T)、Connexin 43、血管内皮生长因子(VEGF)、因子Ⅷ检测。结果对照组的心肌细胞凋亡指数为225.28±125.41,显著高于实验组的78.57±40.29(P〈0.05)。两组Troponin T和Connexin 43均为阴性。实验组的VEGF和因子Ⅷ呈阳性表达。结论骨髓间质干细胞移植对心肌梗死后促进VEGF分泌和因子Ⅷ表达,可减少心肌细胞凋亡,改善心脏功能。Objective To investigate the impact of mesenchymal stem cells implantation on myocardial cell apoptosis in myocardial infarction SD rats. Methods Acute myocardial infarction was made by ligation of the left anterior descending coronary artery in SD rats. After 30 minutes, in the experimental group, 3×10^6 marrow mesenchymal stem cells labelled with Dil were injected into the ischemic myocardium; in the control group, 100μL normal saline were injected at the comparative same site. After 4-6 weeks, TUNEL and immunohistochemical technology were used to detect Troponin T,Connexin 43, VEGF, Ⅷ in situ to estimate the myocardial cell apoptosis within the heart tissue. Results Apoptosis index was 225.28 ± 125.41 in the control group versus 78.57 ± 40.29 in the experimental group (P〈0.05). Troponin T and Connexin 43 were both negative in the two groups, and VEGF,Ⅷ factor was positive in the experimental group. Conclusion Implatation of marrow mesenchymal stem cells into the infarcted myocardum enhances secretion of VEGF and expression of Ⅷ factor, enhancing improvement of cardiac function and decreasing myocardial cell apoptosis.
