摘 要:目的建立测定小鼠血浆中冰片和尼莫地平的气相色谱-质谱(GC-MSn)定量分析方法。分析两药合用后尼莫地平在小鼠体内的药动学变化。方法血浆样品经乙酸乙酯提取,采用选择性离子抽提和二级MS/MS质谱分别监测冰片和尼莫地平的血药浓度。结果本法可同时测定血浆中2种相对分子质量差异较大的药物质量浓度,冰片在0.01~10 mg.L-1,尼莫地平在0.01~0.5 mg.L-1内,峰面积与其质量浓度呈良好的线性关系。最低检测限分别为5.0,2.5 mg.L-1,日内RSD分别为4.6%~6.7%,2.5%~5.2%;日间RSD分别为2.3%~9.9%,5.1%~6.6%(n=5)。结论该方法灵敏,简便,准确,适用于冰片和尼莫地平配伍应用的药动学研究。尼莫地平加冰片合用的动力学研究表明,冰片可以改变尼莫地平在体内的分布过程。OBJECTIVE To establish a method for simultaneous determination of borncol and nimodipine in mice plasma by GC- MS^n. METHODS The plasma sample was extracted with ethyl acetate, borneol and nimodipine were determined by means of selected ion extraction and MS2, respectively. RESULTS The concentrations of the different compounds were simultaneously determined. The calibration curves were linear in the range of 0.01 - 10 mg· L^- 1 for borneol, 0.01 - 0.5 mg· L^- 1 for nimodipine, respectively. The limits of detection were 5.0 and 2.5 mg· L^- 1, respectively. The RSDs of Intra-day validation were 4.6 % - 6.7% ,2.5% - 5.2%, and the RSDs of inter-day validation were 2.3 % - 9.9 %, 5.1% - 6.6 % ( n = 5) for bomed and nimodipine, respectively. CONCLUSION This methods is sensitive, simple, accurate, which could be used to determine simultaneously the concentrations of borneol and nimodipine in mice plasma and study their pharmacokinetics.
