摘  要:目的 探讨经液氮保存的同种带支架瓣膜的制作方法.方法 将制作完成的瓣架经液氮保存1个月后复温,进行几何形态检测(6枚).涤纶包布及缝线经液氮保存1~3个月后复温,检测机械强度变化(6例).采集同种带瓣主动脉和肺动脉,缝于瓣架上,缝制完成后打水观察瓣叶的闭合情况,初步粗略估算Rb/Rc、H/Rc和α值.将制作完成的瓣膜灭菌培养后,程控降温,液氮保存.结果 瓣架直径在液氮冷冻前为(21.1±0.06)mm,冷冻后为(21.1±0.05)mm,P>0.05;瓣架高度冷冻前为(14.03±0.02)mm,冷冻1个月后为(14.04±0.03)mm,P>0.05;几何形态未发生明显变化.涤纶包布经液氮冷冻前纤维方向拉力强度纵向为(19.05±1.64)Mpa,冷冻1个月、3个月后分别为(17.29±1.79)Mpa、(18.23±1.6)Mpa(P>0.05);横向纤维冷冻前为(18.16±1.16)Mpa,冷冻后分别为(16.81±0.97)Mpa、(17.46±1.54)Mpa(P>0.05);包布物理强度未发生明显变化.缝线经液氮冷冻前拉力强度为(163.99±7.83)Mpa,冷冻1个月、3个月后分别为(168.88±6.28)Mpa、(168.74±1.85)Mpa,P>0.05;缝线物理强度未发生明显变化.初步估算Rb/Rc值为1.2左右,H/Rc值为1.4左右,α值为10°左右,瓣架构型均符合制作标准.结论 经液氮保存同种带支架瓣膜的材料选择、设计和制作方法均达到了人工瓣膜的技术要求.Objective To establish the method of the stented homograft valve preserved by liquid nitrogen. Methods The valve frames (n=6) were put into liquid nitrogen, and preserved for one month. After thawing, the geometric shape of frame was measured. The dacron cloth and thread(n = 6) used to fabricate the stented homngraft valve were put into liquid nitrogen, and cyropreserved in one to three months. After thawing, we measured the mechanical strength. The fresh homograft valve was trimmed according to shape of the frame and sewed on the stent. Quality of the prosthetic valves were examined by injecting water before sterilizing, Rb/ Rc, H/Rc and awere estimated. After sterilizing and culture, the prosthese were frozen and transferred into a nitrogen storage refrigerator. Results The results revealed the diameter before preservation was (21.1± 0.06)mm, andafterwas(21.1±0.05) mm(P〉 0.05), the height of before preservation was ( 14.03 ± 0.02)mm, and after preservation was ( 14.04 ± 0.03) mm( P 〉 0.05). The geometric shape of frame was not changed. Our outcomes showed the tension along longitudinal fiber of dacron was ( 19.05 ± 1.64) MPa, and after preservation 1 or 3 months were ( 17.29 ± 1.79) MPa ( P 〉 0.05) and ( 18.23 ± 1.6) MPa ( P 〉 0.05) respectively. The tension along to lateral fiber of Dacron was ( 18.16 ± 1.16) MPa, after preservation were ( 16.81 ± 0.97) MPa ( P 〉 0.05) and (17.46 ± 1.54) MPa (P 〉 0.05 ). The tension of thread before preservation was (163.99 ± 7.83 ) MPa, after preservation were ( 168.88 ± 6.28) MPa ( P 〉 0.05) and ( 168.74 ± 1.85) MPa ( P 〉 0.05). The mechanical strength of dacron and thread were not significantly changed. Rb/Rc was approximately 1.2, H/Rc was approximately 1.4 and awas approximately 10. These parameters accord with standard of prosthetic valve design. Conclusion The stented homograft valve can be cyropreserved by liquid nitrogen and accord with standard of
