摘 要:目的研究金雀黄素对人乳腺癌细胞血管内皮生长因子表达的影响。方法选用两种不同的人乳腺癌细胞株MCF-7和MDA-MB-231体外培养,VEGF免疫组化及原位杂交染色,并用图像分析仪进行定量分析。结果两种人乳腺癌细胞均可转录VEGF-mRNA,并在细胞质中翻译合成相应的蛋白。经金雀黄素处理后两种细胞表达的VEGF都明显减少。结论金雀黄素能通过下调VEGF-mRNA水平,减少VEGF的蛋白表达。Aim To study the effect of genistein on expression of VEGF in human breast cancer cell lines. Methods Two kinds of human breast cancer cell lines ( FCM-7 and MDA-MB-231 ) was cultured in vitro. Immunohistochemistry and in situ hybridization was used to determine the expression of VEGF-mRNA and VEGF protein. The staining results were analyzed using image analysis system. Results VEGF-mRNA was found in these two kinds of cell lines, and corresponding protein was expressed in cytoplasm. Expression of VEGF was significantly lowered in these two kinds of cell lines after treated with genistein. Conclusion Genistein can reduce the expression of VEGF through the down-regulation of VEGF mRNA.
