阿仑瞵酸钠对去势大鼠骨折愈合的影响

摘　　要：[目的]研究阿仑瞵酸钠对骨质疏松性骨折愈合过程的影响。[方法]本研究通过切除大鼠双侧卵巢建立骨质疏松模型,并人为造成后肢骨折后,予以阿仑瞵酸钠灌胃,观察骨折愈合过程中骨痂的组织学、骨密度变化,并用免疫组化方法观察TGF-β1、碱性成纤维细胞生长因子、骨形态发生蛋白-2、胰岛素样生长因子的变化。[结果]X线、组织学显示两组骨折同时愈合。两组TGF-β1、碱性成纤维细胞生长因子、骨形态发生蛋白-2、胰岛素样生长因子含量在14-42 d均出现高峰,经过统计学分析,两组差异无统计学意义。骨密度显示自42 d开始实验组与对照组骨折部骨密度有较明显差别。[结论]阿仑瞵酸钠可使钙离子沉积于软骨痂,提高骨密度,对于预防骨质疏松骨折患者发生再骨折是一种良好的方法。但阿仑瞵酸钠对于骨质疏松骨折愈合的过程,既无延缓作用也无促进作用。[Objective] To study the effect of alendronate on osteoporotic fracture healing. [Methods] Models of osteoporosis were set up by removing beth ovaries of rats, and fracture of hind limb was made on the rats. Then alendronate were given to the rats. Histomorphological changes and bone mineral density were observed during fracture healing. Meanwhile the expression and distribution of transforming growth factor beta 1 （TGF-β1）, basic fibroblast growth factor （bFGF）, bone morphogenetic protein-2 （BMP-2） and insulin-like growth factor （IGF） were observed. [Results] X- ray and histomorphology showed that the fracture healed at TGF -β1, bFGF, BMP-2 and IGF appeared at 14 - 42 day. the same time in the two groups. The peak expression of There was no statistical difference between the two groups. The bone mineral density examination showed that the density of experimental group was higher than control group. [Conclusion] Alendronate may cause the deposition of calcium on callus, increase the bone mineral density. It is a good method to prevent the osteoporotic fracture from fracture again. But alendronate neither promote nor delay the healing of osteoporotic fracture.
