摘  要:目的探讨血管内皮生长因子(VEGF)基因疗法对血管吻合口急性狭窄的预防作用。方法切断兔颈总动脉,进行原位端端吻合,向吻合口血管腔内注入真核表达质粒pcDNA_3/VEGF_(165)(治疗组)和真核载体pcDNA_3溶液(对照组),使之在腔内滞留30 min,应用逆转录-聚合酶链反应(RT-PCR)和蛋白印记法检测吻合口组织的VEGF_(165)基因和其蛋白的表达,同时进行吻合口病理形态学检测和同位素Cr-51标记的血小板计数。结果实验组与对照组术后1周V EGF_(165) RNA和其蛋白表达分别为0.326±0.095,0.137±0.033和0.517±0.140,0.248±0.062差异有统计学意义。治疗组吻合口周围同位素Cr-51标记的血小板数目和吻合口血栓出现率均明显少于对照组。结论将外源性VEGF_(165)基因转入吻合口血管壁中可减少吻合口血小板沉积和黏附,有效地防止术后血管急性狭窄。Objective To study the possibility of preventive effec of gene therapy with VEGF165 gene on the formation of thrombi and intimal hyperplasia of vasvular anastomotic site. Methods End to end anastomoses were performed in rabbit carotid arteries. Replication-defective recombinant pcDNA3/VEGF165 (treatment group) or pcDNA3 (control group) were injected into anastomotic vascular lumen for 30 min. By means of RT-PCR and Western blot assay,the number detection of Cr-51 labelled platelets accumulated at anastomotic sites, pathological study, the following results were obtained. Results The VEGF165mRNA and VEGF in treatment group were sig:,feantly higher than those in control group, The number of platelets accumulated at the anastomotic sites was significantly lower and thrombosis rate fewer in the treatment group than that in the control group ( P 〈 0.01 ). Conclusion When pcDNA3/VEGF165 were transferred into anastomotic vasvular wall,the foreign gene can produce VEGF at anastomotic sites and effectively prevent the formation of thrombosis and intimal hyperplasia.
