摘  要:目的:研究锌原卟啉对大鼠高热惊厥脑损伤CO变化的影响及作用机制。方法:21日龄W istar大鼠,随机分为对照组、高热未惊厥组、FC组及Znpp治疗组。采用热水浴诱导FC大鼠动物模型和Znpp治疗模型。用分光光度计检测各组大鼠脑组织匀浆和血浆CO含量,用HE染色观察大鼠海马神经元形态学改变。结果:FC组血浆CO含量明显升高,与对照组、高热未惊厥组及Znpp治疗组比较,有显著性差异(P<0.01),Znpp治疗组血浆CO含量升高与高热未惊厥组比较,亦有显著性差异(P<0.05);FC组脑组织匀浆CO含量增高,与对照组、高热未惊厥组及Znpp治疗组比较有显著性差异(P<0.01),Znpp治疗组脑组织匀浆CO含量升高,与对照组、高热未惊厥组比较有显著性差异(P<0.01),高热未惊厥组与对照组比较差异无显著性(P>0.05);结论:FC能够引起脑损伤且CO水平增高,Znpp能够使CO水平下降,对脑损伤有保护作用。Objective:The main purpose of the present study was to explore the effect of Znpp on the carbon monoxide(CO) change and the brain damage caused by febrile convulsion ( FC )in rat. Methods: FC in rats was induced by hot water bath, ten times, once every 2 days. 21- day Wistar rats were randomly divided into two groups ,The latter was further divided into none FC group (n= 16) ,FC group(n =20)and Znpp treatment groups(n=15). HE-staining was used to detect the morphologic changes of hippocampal neurons. The indirect CO production in the brain and plasma was detected by the spectrophotometer. Results: The brain tissue homogenate and plasma CO concentration were significantly increased in FC group as compared with that in normal group,none FC group and Znpp treatment group (P 〈 0.01) . The expression of HO-1 mRNA was significantly increased in the hippocampal CA1,CA3 region and dentate gyrus in FC group as compared with that in normal group, none FC group and Znpp treatment group. Conclusions:Recurrent FC in rats may cause brain damage and FC in rats can cause the CO increase. Znpp inhibits the CO to have a protective effect on FC brain damage.
