摘  要:目的探讨慢性阻塞性肺病(COPD)肺内细胞凋亡与增殖的关系。方法22只大鼠分为正常组、对照组、慢性支气管炎组、慢性支气管炎+肺气肿组,采用免疫组化法及原位缺口末端DNA碎片标记技术(TUNEL法)检测各组大鼠肺内细胞凋亡与增殖。结果慢性支气管炎、慢性支气管炎+肺气肿组的细胞凋亡指数、增殖指数均有不同程度增高,与正常组相比较差异有统计学意义。凋亡细胞主要分布在气管上皮细胞、肺泡上皮细胞、血管内皮细胞以及气管周围少许炎细胞,增殖细胞绝大多数为位于气管上皮细胞、气管周围炎性细胞。结论慢性支气管炎、慢性支气管炎+肺气肿组中凋亡和增殖均有不同程度增高,增殖增高水平高于凋亡,细胞增殖与凋亡的失衡可能在COPD发病机理中起到一定的作用。Objective To study the relation of apoptosis and proliferation in rats with chronic obstructive pulmonary disease ((33PD). Methods Twenty-two Wistar rats were randomly divided into four groups: normal group, control group, chronic bronchitis (CB) group,CB and emphysema group. Immuocytochemistry technique was used for the detection of proliferative cellsand situ end-labeling technique was used for the detection of apoptotic cells in the lung. Results Both proliferative cells and apoptotic cells were found in every group. Compared with normal group, the proliferation index and apoptosis index were significantly increased to varied extent in model groups. In the lung tissue of CB and emphysema group, the proliferative antigen were predominanfly localized in bronchial epithelial cells, alveolar epithelial cells, small pulmonary vessels and some inflammation cells infiltrating around the airways, and the apoptotic cells were predominantly localized in bronchial epithelialcells and inflammation cells infiltrating around the airways. Conclusion Both proliferation and apoptosis are found in CB group, CB and emphysema group. The changes of proliferative cells are significantly higher than those of apoptotic cells. Theabnormal increased expression of apoptosis and proliferation may contribute to the development in COPD rats.
