摘  要:目的研究中药“十八反”中大戟与甘草配伍对大鼠肝功能和肝脏微粒体中CYP3A2在mRNA水平、蛋白表达及酶活性方面的影响。方法采用RT-PCR技术检测大鼠肝脏CYP3A2mRNA表达水平;利用Western-blot技术检测大鼠肝脏CYP3A2蛋白的表达;采用红霉素N-去甲基反应评价CYP3A2酶活性。结果大戟和甘草合用对大鼠肝功能损害最为严重;在mRNA水平、蛋白表达、酶活性方面,大戟组均高于空白组及其他各单用组。结论大戟、甘草可能存在基于药物代谢酶机理的相反作用,但需进一步结合代谢研究加以综合分析,阐明相反作用的机理。Objective:To study the influence on the rats' liver factor and the mRNA,protein expression level and activity of cytochrome P450 3A2 (CYP3A2) by combined use of peking euphorbia and licorice root in microsomal.Methods:The mRNA expression level of CYP3A2 were detected by semi-quantitative reverse transcriptase-polymerase chain reaction(RT-PCR).The protein expression level of CYP3A2 were detected by Western-blot technology.The activity of CYP3A2 were quanititated by erythromycin demethylation Method.Results:The damage of liver factor in combined group were obviously while the other groups were not.The expression of mRNA and protein of CYP3A2 were higher than the other groups as well as its activity in peking euphorbia group.Conclusion:peking euphorbia with may licorice root occur drug-drug interaction based on CYP3A2.To elucidate the drug-drug interaction,it needs further analysis and metabolism research.
