摘 要:目的探讨前列腺素E1对肺过度膨胀所致损伤的保护作用及作用机制。方法杂种犬16只,随机分为对照组和实验组,建立肺过度膨胀动物模型。对照组:保持肺过度膨胀6h;实验组:保持肺过度膨胀6h,并以30ng/(kg·min)的剂量给予前列腺素E1,6h后取标本进行肺组织湿/干比、支气管肺泡灌洗液中蛋白含量的检测,并进行肺组织光镜和电镜的观察,观察肺损伤的变化情况。结果对照组肺组织湿干比、支气管肺泡灌洗液中蛋白含量(BALF)明显高于实验组(P<0.05);肺组织学检查:实验组肺损伤程度明显轻于对照组。结论前列腺素E1对犬肺过度膨胀所致损伤有明显的保护作用,其作用机制与抑制中性粒细胞活性,降低中性粒细胞、血小板聚集等有关。Objective To explore the protective effect of prostaglandin E1 ( PGE1 ) on lung and its possible mechanism during lung hyperinflation injury. Methods Sixteen mongrels were randomly divided into control group ( C ) and PGE-n = 1 group( P ) and lung hyperinflation canine model was established. Lung hyperinflation was kept in control group for 6 hours, and on the basis of the former the PGE1 group was given 30 ng/kg/min PGEl. The changes of lung tissue wet weight/dry weight ratio ( W/D ) and total protein content of bronchoalveolar lavage fluid ( BALF ) were measured after 6 hours; the histological appearances of the lung tissue and the pulmonary vascular were observed under histology and electron microscopy. Results W/D and total protein content of BALF were significantly higher in control group than in PGE1 group( P 〈 0.05 ) ; histological examination of lung tissue demonstrated that the degree of lung injury was worsen in PGE1group than in control group. Conclusion PGE1 can significantly attenuate the lung hyperinflation injury, which protective mechanism may be that PGE1 can inhibit aggregation of neurotrophilic granulocyte and platelet in the lung.
