摘  要:目的建立鬼臼毒素固体脂质纳米粒含量的高效液相色谱测定方法。方法以ODS-C18(4.6mm×250mm,5μm)为分析柱,以甲醇-水-冰醋酸(59:41:0,6)为流动相,流速0.8mL·min^-1,检测波长为285nm,进样量20μL,柱温26℃。结果当鬼臼毒素浓度在1.6~80.0μg·mL^-1。范围内时,峰面积与浓度之间呈良好的线性关系(r=0.9999,n=5),高、重、低3个剂量的平均回收率分别为99.09%,99.88%,100.92%。日内和日间精密度RSD分别为0.39%,0.42%,3批鬼臼毒素固体脂质体中鬼臼毒素的含量分别为94.89%,94.76%,97.24%。结论该方法准确,简便,灵敏,重现性好,适合测定鬼臼毒素的含量。Objective To establish the HPLC method for the determination of podophyllotoxin in pod-sin. Methods ODS-C18 (4.6 mm×250 mm, 5 μm) was used as the stationary phase. The mobile phase was the solution containing methanol-water-glacial acetic acid (59 : 41 : 0.6); flow rate: 0.8 mL·min^-1; wavelength of UV detection: 285 nm, sample: 20 μL and temperature of column: 26 ℃. Results The calibration curves was linear in the concentrations of 1.6~80.0 μg·L^-1 (r =0.999 9, n = 5). The average recovery of three doses was 99.09%, 99.88%, 100.92%; the contents of podophyllotoxin in 3 batches pod-sin were 94.89%, 94.76%, 97.24%, respectively. Conclusion The method is simple, accurate, rapid and reproducible. It is suitable to assay the content of the drug.
