摘 要:目的研究去窦弓神经(SAD)大鼠血浆血管紧张素Ⅱ和内皮素1水平的明亮-黑暗周期变化模式,探讨其与血压异常波动的联系。方法72只12周龄SpragueDawley大鼠随机分为手术去除窦弓神经组和假手术组,各36只,术后4周测定两组大鼠24h血压和血压波动性(BPV)。在光照-黑暗交替(LD)光制下适应10d后,再将两组大鼠各自随机分为6组,每组各6只,明亮-黑暗周期内每隔4h采集1组血标本,采用放射免疫分析技术测定不同人工明亮-昼夜周期时点(ZT)血浆血管紧张素Ⅱ(AngⅡ)和内皮素1(ET1)水平。结果SAD大鼠术后4周,24h平均收缩压(SBP)和舒张压(DBP)与假手术对照组比较无显著差异,24hSBPV和DBPV均显著高于对照组大鼠[(SBPV:13.8±4.2vs对照组:6.4±1.1),(DBPV:11.5±3.1vs对照组:5.3±0.8)mmHg,P均<0.01]。血浆AngⅡ和ET1水平均呈现明显的明亮-黑暗周期节律性变化,假手术大鼠血浆AngⅡ水平呈明亮-黑暗周期双峰模式(F=2.82,P<0.05,ANOVA),光照期峰值位于ZT8(776.9±189.5)pg/mL,黑暗期峰值位于ZT16(689.5±199.6)pg/mL;血浆ET1变化呈昼(明亮)低夜(黑暗)高模式(F=2.749,P<0.05,ANOVA),其峰值和谷值分别位于ZT20(峰值:193.7±28.5)pg/mL和ZT4(谷值:131.1±26.5)pg/mL;而SAD大鼠血浆AngⅡ为昼(明亮)高夜(黑暗)低模式(F=5.29,P<0.01,ANOVA),其峰值和谷值分别位于ZT4(峰值:1014.6±130.9)pg/mL和ZT16(谷值:220.4±147.6)pg/mL;血浆ET1变化呈明亮-黑暗周期双峰模式(F=2.68,P<0.05,ANOVA),峰值分别位于ZT4(峰值:228.3±54.0)pg/mL和ZT20(谷值:213.7±54.6)pg/mL。结论SAD大鼠血浆血管紧张素Ⅱ和内皮素1的明亮-黑暗周期分泌节律异常,并对血压波动性的调节可能有一定作用。Objective To investigate the changes in plasma levels of angiotensin Ⅱ , endothelin-1 and blood pressure fluctuation during light-dark aherative period in sinoaortic denervated (SAD) rats. Methods Seventy-two male Sprague-Dawley rats underwent SAD or sham operation at the age of 12 weeks. Twenty-four-hour blood pressure and blood pressure variability (BPV) were measured in conscious, unrestrained rats 4 weeks after operation. Rats were housed in a 12 h light/12 h dark cycle (LD 12: 12) for at least 10 days, then the blood samples were taken every 4 h. The plasma levels of angiotensin Ⅱ and endothelin-1 were assayed by radioimmunoassay (RIA). Results No significant difference in SBP or DBP between SAD and sham-operated rats was found. Compared with sham-operated rats, SAD rats had greater SBPV and DBPV over 24 hours ( SBPV: 13.8 ± 4.2 vs control: 6.4± 1.1, DBPV: 11.5 ± 3.1 vs control: 5.3±0.8 mm Hg, all P〈0.01 ). Diurnal variations of plasma angiotensin Ⅱ and endothelin-1 in SAD and sham-operated rats were significant (all P〈0.05, ANOVA). In sham-operated rats, a bimodal pattern of plasma angiotensin Ⅱ was demonstrated (F= 2. 817, P〈0.05, ANOVA ) with peak concentration at ZT8 (776.9±189.5 pg/mL) in the light phase and ZT16 ( 689.5±199.6 pg/mL) in the dark phase. Plasma endothelin-1 level was increased in the dark phase while was lowered in the light phase (F =2. 749, P〈0.05, ANOVA). Instead of peak in dark phase Ang Ⅱ was lowered without changes in Ang Ⅱ peak in light phase in SAD rats (F=5.29, P〈0.01, ANOVA ). Plasma endothelin-1 level exhibited a bimodal profiles (F=2. 679, P〈0.05, ANOVA ). Conehtsion The diurnal change pattern of plasma angiotensin Ⅱ and endothelin-1 were altered in SAD rats. The circadian variations in plasma Ang Ⅱ and endothelin might play role in the modulation of blood pressure variability.
