人血浆中盐酸莫雷西嗪的高效液相色谱法测定及其药物动力学研究

摘  要:目的 建立测定人血浆中莫雷西嗪浓度的高效液相色谱法。方法 色谱柱:HypersilODS-2(150nm×4.6mm,5μm);流动相:甲醇:水:三乙胺(70:30:0.4),v/v/v,pH6.8);检测波长:268nm;以氯氮平作为内标,测定人血浆中的莫雷西嗪药物浓度。结果 莫雷西嗪浓度线性范围为0.04~4mg/L,回归方程Y=11.42X+1.441,r=0.9992;回收率96、1%,日内和日间精密度(RSD)分别为4.7%和5.3%,整个测定时间8min左右。结论 该方法简便、快速、准确,适用于临床上莫雷西嗪药物动力学的研究。Objective To estabish a HPLC method for determining the concentration of moricizine in human plasma. Methods The plasma concentrations of moricizine were determined by HPLC method. The assay was performed on the Hypersil ODS-2 column with the mobile phase consisted of methanol-water-triethylamine (70:30:0.4, v/v/v, pH 6.8). The UV detector wavelength was set at 268 nm,and the clozapine was taken as the internal standard. Results The calibration curve was linear in the range of 0.04 ~ 4 mg/L with a regressive equation of Y=11.42 X + 1.441, r = 0. 999 2. The recovery of moricizine was 96.1% ; the relative standard deviations were 4.7 % (intra day) and 5.3 % (inter day) respectively. Conclusion The method is shown to be, conveniem, accurate, rapid,and suitable for detemlining moricizine in the human plasma and pharmacokinetic studies of moricizine.
