MGEA5基因单核苷酸多态性与中国北方汉族人群2型糖尿病的相关性研究

摘  要:目的研究M GEA 5基因单核苷酸多态性与中国北方汉族人群2型糖尿病(T 2DM)的相关性。方法实验分糖尿病组(DM)和正常对照组(NC),用酚/氯仿法提取外周血DNA行PCR扩增,扩增产物用1.5%琼脂糖电泳,分离纯化目的条带后直接测序。结果M GEA 5基因的内含子10中,SNP 5-14位点等位基因为A/T,携带等位基因T的个体DM发病率比携带等位基因AA的个体显著增加,该等位基因T频率在两组间的差异有统计学意义(P<0.01)。结论M GEA 5-14等位基因可能是中国北方汉族T 2DM的主要易感基因位点。[Objective] To investigate the correlation between single nucleotide polymorpbism MGEAS-14 and type 2 diabetic mellitus in Han people of Northern China. [Methods] 155 diabetic mellitus patients and 126 normal control subjects were involved in the experiment. Phenol/chloroform method was used to extract the DNA from the peripheral blood. PCR method was used to amplify the aimed gene fragment, the amplified product was electropboresed on the agarose gel with ethidium bromide, and the aimed strap was purified and sequenced. [Results] The allele snpS-14 in the MGEA5 gene within the 10th intron was A/T, subjects carrying the T allele suffer an increased incidence of diabetic mellitus than those carrying the AA allele. The allele T frequency in the diabetic mellitus group was significantly higher than that in the normal control group (P 0.01). [Conclusion] The allele gene MGEAS-14 might be the major susceptibility gene that responsible for the type 2 diabetic mellitus of Han people of Northern China.
