摘 要:目的探讨肝细胞生长因子(hepatocyte growth factor,HGF)在妊娠期高血压疾病(hypertensive disorder complicating pregnancy,HDCP)妇女胎盘组织中的表达及对胎盘绒毛滋养细胞凋亡的影响。方法选取不同程度HDCP晚孕孕妇34例及正常晚孕孕妇30例。采用逆转录聚合酶链反应(RT-PCR)的方法检测其胎盘组织中HGF和Fas mRNA表达量。结果(1)与对照组(0.67±0.19)相比,妊娠期高血压组HGF mRNA表达无显著差异(0.65±0.06,P>0.05),轻度子前期、重度子前期和子组HGF mRNA表达显著降低(0.43±0.12,0.38±0.09和0.19±0.17,P<0.05),且随病情加重,表达强度逐渐降低;(2)轻度子前期、重度子前期和子组FasmRNA与对照组相比,表达显著升高(1.58±0.26,2.96±0.14,5.98±1.17和1.01±0.36,P<0.05),且随病情加重,表达强度逐渐增加;(3)胎盘组织中HGF和Fas表达呈负相关(r=-0.618,P<0.05)。结论胎盘间质细胞表达的HGF具有抑制由Fas介导的滋养细胞凋亡作用。患HDCP孕妇胎盘HGF表达下降可能是引起滋养细胞凋亡的原因之一。Objective To investigate the expression of hepatocyte growth factor(HGF) in placentae of women with hypertensive disorder complicating pregnancy (HDCP), and the effect of HGF on apoptosis of trophoblasts. Methods RT-PCR were used to determine the expression of HGF and Fas in 30 cases of uncomplicated pregnancy and 34 cases of HDCP. Results The expression of both HGF mRNA and Fas mRNA in the mild preeclampsia group, severe preeclampsia group and eclampsia group was significantly lower or higher than that of the control (HGF mRNA: 0.43±0. 12, 0. 38± 0. 09 and 0.19±0.17 vs 0.67±0.19, Fas mRNA.. 1.58±0.26, 2.96±0.14 and 5.98±1.17 vs 1.01 ±0. 36, P〈0.05), while no difference was found betweenin the gestational hypertension group and the control group (0.65±0.06 and 1.13±0.20, P〉0. 05). Decreased HGF mRNA or increased Fas mRNA was found along with the progress of HDCP. Negative correlation was shown between the expression of HGF and Fas (r= -0. 618, P〈0.05). Conclusions HGF secreted by mesenchyme in the placenta inhibits the apoptosis mediated by Fas , and the reduced expression of HGF in HDCP may be responsible for the apoptosis of the trophoblasts.
