摘 要:目的比较生育与不育男性精子顶体反应释放的顶体酶活性,以此来评价精子顶体反应。方法30例不育男性和10例健康生育男性精液,用Percoll密度梯度分离法优选精子,FITC-PSA荧光染色法分析AR;用分光光度比色法测定顶体酶活性。结果少精子症组、弱精子症组和畸形精子症组精子顶体反应率和顶体反应释放的顶体酶活性均明显低于生育组(P<0.05),前3组中顶体反应率和顶体反应释放的顶体酶活性以畸形精子症组最低,但不育各组间比较均没有明显差异。将顶体反应释放的顶体酶活性与其相应的顶体反应率进行相关分析,两者存在明显正相关关系(P<0.001)。结论用顶体反应释放的顶体酶活性可以评价精子顶体反应功能。Objective To investigate the acrosin activity as an indicator to evaluate the function of acrosome reaction. Methods Human semen was collected from 30 infertile men and 10 fertile donors with normal semen parameters. Specimens were allowed to liquify at 37 ℃, and were prepared by Percoll-gradient treatment. Spermatozoa were recovered from the upper layer. Acrosome reaction was analyzed using FITC-PSA staining. Acrosin activity was analyzed with colorimetry by spectrocolorimeter. Results Progesterone could induce acrosome reaction. The percentage of acrosome reacted sperms and released acrosin activity of oligozoospermic ; asthenozoospermic and teratozoospermic groups were all lower than those of fertile groups ( P 〈 0. 05 ). The percentage of acrosome reacted sperm and released acrosin activity of teratozoospermic group was the lowest among three infertile groups. But there was no difference among oligozoospermic, asthenozoospermic and teratozoospermic group. There was a markedly positive relationship between the percentage of acrosome reacted sperm and released acrosin activity( r = 0.938, P 〈 0.001 ). Conclusion Released acrosin activity of acrosome reacted sperm can function as an indicator to evaluate the function of acrosome reaction and can be one of routine test for evaluation of male eugenesis.
