摘　　要：目的研制检测麻疹IgM抗体的酶联免疫（EIA）试剂盒。方法以经超滤、PEG沉淀和蔗糖密度梯度超速离心纯化后的沪191株麻疹病毒为包被抗原，经试验确定EIA检测的最佳反应条件，最终建立麻疹病毒IgM EIA检测试剂盒，并与HUMAN麻疹IgM EIA检测试剂盒作比较。结果经Western—blot证明，纯化后沪191株麻疹病毒特异性符合要求。以该纯化抗原制备的EIA检测试剂盒具有良好的重复性（CV为4．76％～11．96％）。4℃放置6个月、57％放置3d，该试剂的灵敏度不变，说明其具有良好的稳定性。与HUMAN试剂盒相比，该试剂盒的灵敏度为94．12％，特异度为92．31％，一致性为93．91％。结论沪191麻疹IgM诊断试剂可以正确反应麻疹病毒的感染情况，可以作为一种可靠、简便、快速诊断麻疹病毒感染的实验室方法。Objective To develop an EIA diagnostic kit in detecting anti- measles IgM.Methods S191 measles virus purified by ultrafihration, PEG precipitation and density gradient uhracentrifugation was immobilized on polystyrene strip. The anti - measles IgM EIA diagnostic kit was established after determining the optimal reaction conditions by a series of tests. The kit was compared with HUMAN antimeasles IgM EIA kit. Results The purity and specificity of the purified S191 measles virus were determined by SDS - PAGE and Western blot. The reproducibility（CV：4.72% - 13.51 % ） of the kit which used S191 as coating antigen to detect anti - measles IgM was excellent, it maintained the same quality even when stored at 4℃ for 6 months or at 37℃ for 3 days. Compared with HUMAN anti - measles IgM EIA kit, the newly- developed kit has a sensitivity of 94.12% and specificity of 92.31%, the agreement rate being 93.91%. Conclusions The anti - measles IgM EIA kit is a reliable, convenient and rapid means for detecting infection of measles virus.
