摘 要:采用液相杂交技术,对二氢埃托啡(DHE)和吗啡(Mor)处理大鼠的中脑,海马和纹状体中μ受体mRNA含量进行测定.大鼠皮下注射DHE3d或Mor14d可分别对DHE或Mor产生耐受.中脑μ受体mRNA水平在DHE或Mor处理3d即下降,海马和纹状体中μ受体mRNA在处理7d或21d后亦开始下降.表明活体DHE或Mor处理可引起大鼠不同脑区μ受体mRNA下调,这种下调可能与DHE或Mor的耐受有关,同时耐受的产生可能还有其它机理.The μ opioid receptor mRNA levels in rat midbrain, hippocampus and striatum after dihydroetor- phine(DHE) and morphine (Mor) treatment were tested by solution hybridization. Rats rendered tolerance to DHE or Mor after 3 d of DHE or 14 d of Mor treatment. Whereas, the μ receptor mRNA level was decreased in midbrain after only 3 d of DHE or Mor treatment. And the μ receptor mRNA levels in hippocampus and in striatum were also decreased after 7 and 21 d of DHE or Mor treatment respectively. These results indicate that down-regulation of μ opioid receptor mRNA can occur in vivo in rat brain regions after DHE or Mor treatment and this down-regulation of μ receptor may be correlated to DHE or Mor tolerance. However, in addition to μ receptor down-regulation, other mechanisms may also be involved in tolerance.
