摘  要:解偶联蛋白3是一种线粒体内膜上的转运蛋白,呵以使氧化过程与ADP磷酸化过程解偶联。本研究分离了猪UCP3基因的外显子2~3和外显子6。经过序列分析,发现猪UCP3基因共有3处框移突变和18个SNP位点。在猪UCP3片段中分别包含一个SmaⅠ和SduⅠ酶切位点。UCP3片段SmaⅠ位点在大白×梅山F2家系中经PCR—RFLP分析,表现为AA、AB和BB3种基因型,A基因频率为0.57,B基因频率为0.43。因此,可以将UCP3基因作为猪分子遗传育种的候选基因。Uncoupling protein 3 (UCP3) was mitochondrial membrane transporters involved in themogenesis, which uncoupled the oxida- tion and phosphorylation of ADP on respiration chain. Exon 2 - 3 and 6 of UCP3 gene were isolated in this study. By DNA sequences analysis, three frame-shift mutation and eighteen SNP were found in UCP3 gene of pigs. There were restriction enzyme sites of Sma Ⅰand Sdu Ⅰ in UCP3 gene of pigs. The characterization of the Sma Ⅰ polymorphism in UCP3 gene was analyzed by PCR-RFLP among "Largewhite x Meisban" F2 group. The genotypes of UCP3 were AA, AB and BB. The frequencies of A allele is 0. 57 and B is 0.43.
