摘  要:目的研究导入血管紧张素Ⅱ受体1反义基因对大鼠高盐饮食诱导的高血压的预防作用。方法首先采用钙磷转染法进行三质粒共转染,包装重组腺相关病毒,后采用Dot-Blot方法检测病毒滴度。选取8周龄雄性SD大鼠分成3组,分别经尾静脉注射重组腺相关病毒和生理盐水,实验组(rAAV-AT1-Antisense),对照组(rAAV-GFP),正常组(0.9%。NaCl);2周后,实验组和对照组给予高盐(8%氯化钠)饲料喂养,正常组继续给予正常饮食喂养,实验周期为12周,实验过程中,前三周每周测量血压一次,三周后,每两周测量血压一次;处死动物后通过半定量RT-PCR检测血管紧张素Ⅱ受体1在主动脉组织的表达情况。结果高盐饮食1周后,对照组大鼠血压开始升高,到第5周时血压达到140 mm Hg,并维持到实验结束,实验结束时达到(142.7±4.5 mmHg);实验组大鼠和正常组大鼠的血压一样在整个实验过程中一直稳定在正常范围,实验结束时维持在(117±3.8)mmHg, (117.8±2.9)mmHg。整个实验过程三组动物并未出现因为病毒载体注入和表达所引起的严重毒副作用。结论血管紧张素Ⅱ受体1反义基因可以预防高盐诱导的高血压,为临床应用反义AT1基因治疗高血压提供了可能。Objective To investigate the effects of rat angiotensin Ⅱ receptor 1 antisense gene on high salt diet induced hypertensive rats' blood pressure. Method The rAAV-D ( ± ) virons containing AT1-Antisense and GFP (as control), respectively, were packed by co-transfections of 293 cells and tittered by dot blots. 18 male SD rats were allocated randomly into three groups: A single tail vein injection of rAAV- D( ± ) -AT1 -Antisense or rAAV-D( ± ) -GFP ( about 2×10^-10 p. f. u/rat. ) or 0.9% NaCl into rats was performed. After 2 weeks, high salt (8% NaCl) with rAAV-D( ± )-AT1-Antisense group, and high salt (8% NaCl) with rAAV-D( ± )-GFP group, normal diet with normal group. A high salt diet (8% NaC1) and normal diet was given for 12 weeks. Blood pressure, the volume of urine and water were observed during the experiment. Result The rAAV- D ( ± )-GFP-treated animals developed significantly higher systolic blood pressure, but the rAAV- D( ± )-AT1 -Antisense treatment attenuated the raise in the blood pressure (142.7 ±4.5mmHg vs. 117 ±3.8mmHg, P 〈0.01) and the hypotensive effect lasted until the end of the experi- mental duration ( 12 weeks). Conclusions rAAV-D( ± ) mediated delivery of AT1-Antisense gene can be an excellent gene remedy for hypertension.
