Effect of Anti-TRX s Gene on Thioredoxin Reduction Characteristics in Seeds of Transgenic Wheat

摘  要:以转反义TRX s基因小麦纯合系为材料,分析了转基因小麦TRX h活性和清蛋白、球蛋白、谷蛋白和麦醇溶蛋白各组分及其还原状态的变化。结果表明,不同成熟期转基因小麦籽粒和胚乳中TRX h活性平均比对照降低76.2%和14.2%,其籽粒中的清蛋白、球蛋白、麦醇溶蛋白和谷蛋白巯基含量平均比对照分别低41%、44%、14%和13%,胚乳中分别比对照低37%、17%、45%和6%,而不同成熟期转基因小麦籽粒和胚乳中4种蛋白组分含量比对照都相应提高。说明导入的反义TRX s基因对小麦TRX h基因表达有显著(P<0.05)的抑制作用,进而影响TRX h对各蛋白组分的还原作用,造成还原态蛋白组分(巯基含量)降低,被动员消耗的蛋白质数量减少,而各蛋白组分含量显著(P<0.05)高于对照。本研究结果为转反义TRX s基因小麦种子发芽延缓和抗穗发芽特性提供了直接的证据。Thioredoxins (TRX) are small ubiquitous proteins, containing a readily reducible disulfide bridge. TRX protein plays a role in a wide variety of physiological processes by acting as general protein disulfide reductases and has been proved to have a function of accelerating seed germination. Anti- TRX s gene transferred to wheat can decrease thioredoxin expression and reduce its activity in wheat, so that the pre-harvest sprouting at mature stage can be slowed down. We have obtained the transgenic wheat ( Triticum aestivum L. ) with anti- TRX s gene, and the T3 and T4 generations of its pure line 00T89. In the present study, the T3 and T4 seeds of 00T89 were planted in both greenhouse and field to investigate TRX h activity, protein components (albumins, gliadins, globulins and glutelins) and free -SH contents in grains and endosperm, with non-transgenic plant as control. The seeds at same panicle position and with same anthesis date were sampled at milk, dough and mature stages, respectively for measurement. The results showed that the TRX h activities in seed and endosperm of transgerfic plant were lower than those of control by 76.2% and 14.2% at different mature periods. The free -SH contents of albumins, gliadins, globulins and glutelins were lower by 37%, 17%, 45% and 6% in seed, and 37%, 17 %, 45 % and 6 % in endosperm, respectively. However, the contents of albumins, gliadins, globulins and glutelins in seeds increased more than those in control at different mature periods. The results suggest that the anti-TRX s inhibites TRX h gene expression, and leads to the decreases of the free -SH content in various proteins and increases the contents of albumins, gliadins, globulins and glutelins, resulting in slower germination in transgenic wheat.
