摘 要:目的探讨内毒素致新生大鼠急性肺损伤与IL-18水平的关系。方法内毒素5mg/kg腹腔注射7日龄SD新生大鼠致急性肺损伤模型,同时设腹腔内注射生理盐水对照组,每组12只。注射后1、4、8h采集血,1、4h采集肺组织,检测血浆,肺组织匀浆IL-18水平。结果内毒素注射后新生大鼠1h,血浆IL18水平就显著上升(6.38±1.62)pg/ml,4h达到高峰(34.28±17.89)pg/ml,8h后明显回落(14.61±2.16)pg/ml,仍高于生理盐水对照组(1.63±1.18)pg/ml;4h组肺组织匀浆IL-18水平(508.96±105.38)pg/ml显著高于1h组(346.08±21.23)pg/ml(P<0.01),且肺组织匀浆IL-18水平高于同时点血浆IL18水平(P<0.01)。结论IL18是一种早期的致炎因子,在急性肺损伤的发生中起重要的作用,可作为急性肺损伤的诊治指标之一。Objective To investigate pro-inflammatory interleukin-18 (IL-18) change and its relation to lipopolysaccharide (LPS) induced acute lung injury (ALI) of neonatal rat. Methods ALI animal model was made by intraperitoneal injection of LPS 5 mg/kg. Enzyme-linked immunosorbent assay (ELISA) was used to determine plasma or lung tissue IL-18 level in neonatal rats 1, 4 and 8 h after injury. Results The plasma IL-18 level increased 1 h after hit with LPS, ( 6. 38 ± 1.62 ) pg/ml, it reached peak level at 4 h, (34.28 ± 17.89) pg/ml, and decreased rapidly at 8 h ( 14. 61 ±2.16) pg/ml. Lung tissue IL-18 level at 4 h increased significantly compared with that at 1 h, (508.96 ± 105. 38 )pg/ml vs (346.08 ±21.23) pg/ml, P 〈0.01. Lung tissue IL-18 level was higher than plasma IL-18 level at the same time, P 〈0. 01. Conclusions IL-18 is an early pro-inflammatory cytokine and it may play an important role in the development of ALI.
