牛膝染色体的制备方法研究

摘　　要：对药用植物牛膝的染色体制备技术进行了初步研究与改进。以牛膝的根尖为试验材料,运用8-羟基喹啉,风油精和变温处理法进行前处理,结合去壁低渗-火焰干燥法中的低渗处理对牛膝染色体标本的制备方法进行了大胆探索。结果表明:采用8-羟基喹啉对牛膝进行前处理结合低渗处理可以获得良好的染色体图像。This study was carried out with root tips of Achyranthes bidentata BI. The treatment methods and times for root tips were studied. With the pre-treatment of 8-hydroxyquiline,fenyoujing and temperature change, the preparation method of chromosomes of Achyranthes bidentata BI was studied by low osmosis treatment after the displacement of cell wall and flame desiccation .The result showed that： a good photo of chromosome can be obtained by using 8-hydroxyquiline to pre-treat Achyranthes bidentata BI combined with low osmosis treatment.
