摘 要:选用抗虫棉品种DP99B,彩色棉品种绿絮1号和棕絮1号,应用现代生物科学技术和手段,研究了棉花早期(萌动期、子叶期、1叶期)热激诱导产生的热激蛋白(HSPs)种类及其含量。结果表明:3个品种3个时期热激处理,处理后41.8 KD热激蛋白含量都增加。此外,DP品种不同时期热激处理后蛋白质含量增加的有3种,含量减少的有1种,含量不变的有3种,含量测不出来的有10种;LV品种热激处理后蛋白质含量增加的有3种,含量减少的有2种,含量不变的有3种,含量测不出来的有8种;ZH品种热激处理后蛋白质含量增加的有6种,含量减少的有2种,含量不变的有2种,含量测不出来的有5种。Selecting one boll-worm-resistant cotton variety (DP) and two colored cotton varieties (LV and ZH), cotton seedling growth was heat shock at different growing periods such as germination stage, cotyledon stage and one-leaf stage. The method of heat shock is from 25℃ directly moves in 42℃, the relative humidity 65% artificial illumination incubator, deal with two hours. The soluble protein in cotyledon was extracted, and heat shock protein was determined with the SDS-PAGE, electrophoresis gel was discriminated by the GDS 7600 gelatin imagery and the analysis system of English UVP Corporation, molecular weight and the content of every protein was surveyed using fitting protein analysis software. This study shows: Synthesis of heat shock protein had 4 kind of expression after heat shock: the content of soluble protein increased, the content reduced, the content was invariable and others couldn't be detected. Three varieties synthesized the protein with molecular weight of 41.8KD and increased in contents by heat shock in different stages. Furthermore, DP increased in the content for three kinds(41.8KD,36.3KD,31.5KD), decreased in the content for one kind(22.9KD), unchanged in contents for three kinds (25.6KD, 24.4KD, 19.0KD)and detected for ten kinds. LV raised in contents three kinds (41.8KD,24.3KD,19.1KD)of HSPs, two kinds (32.1KD,20.9KD)reduction, three kinds (31.5KD,25.3KD,23. 5KD)homology and eight kinds detection. ZH raised in contents six kinds (73. 3KD, 41. 8KD, 32. 1KD, 31. 5KD, 25. 2KD, 23. 1KD) of HSPs, two kinds.(42.6KD,19.1KD)reduction, two kinds(71.5KD,25.2KD)homology and five kinds detection. The possible reason for gain of protein content is that the HSPs combined sequence has a stronger tolerant ability for the high temperature, the synthesis protein is necessary for maintenance; these protein combined sequences are extremely sensitive to the high temperature, which the content of protein is reduction and invariable, and the synthesis is blocked
