摘 要:目的研究压力超负荷致心肌肥厚模型大鼠心脏结构的变化特点及小檗碱对此的干预作用。方法采用腹主动脉结扎法制备压力超负荷型心肌肥厚大鼠模型,造模后2周开始给药,连续10周。以左心室前壁厚度、左心宣腔横切面积、组织羟脯氨酸含量以及心肌间质胶原容积分数(ICVF)等作为心肌肥大和重塑的指标,观察药物的作用。结果结扎腹主动脉12周后,模型组动物心室前壁厚度、心室腔横切面积及左心室壁横切面积均较假手术组动物增加,并显示同心性肥大的特点。且左心室组织羟脯氨酸含量及ICVF均比假手术对照组明显增加,表明心肌间质胶原沉积是压力超负荷致心肌肥厚和重塑的特征之一。小檗碱5、10、20 mg/kg和阳性对照药物卡托普利45 mg/kg连续给药10周对上述变化有一定改善作用。结论小檗碱可能通过抑制心肌间质纤维化而改善心室重构。Objective To study the characteristics of heart structure changes in rat model with hypertrophic cardiomyopathy (HCM) induced by pressure-overload, and the interference of berberine. Method The rat model with HCM was established by using the ligature of ventral aorta and then given berberine for 10 weeks from 2nd week. The rat left ventricular front wall thickness, cross-sectional area of left ventricular, hydroxyproline (Hyp) content and interstitial collagen volume fraction (ICVF) were taken as the indexes of myocardial hypertrophy and ventricular remodeling. Result In the model group the rat left ventricular front wall thickness, cross-sectional areas of left ventricular and left ventricular wall were all increased compared with those of the sham-operated group, and showed a characteristics of concentric hypertrophy. The Hyp content and ICVF were increased in left ventricular tissue significantly in the model group compared with those in the sham-operated group. The result showed that one of the characteristics of HCM and ventricular remodeling induced by pressure-overload was collagen sludging of myocardial interstitium. The administration of berberin (5, 10 and 20 mg/kg respectively ) and positive control drug-Captopril (45 mg/kg) for 10 weeks continually had relieving effects on above-mentioned changes Conclusion Berberin may relieve the ventricular remodeling through inhibiting the fibrosis of myocardial interstitium.
