摘 要:利用荧光光谱、紫外-可见吸收光谱、圆二色谱、琼脂糖凝胶电泳方法研究了小檗碱(berberine)和溴化乙锭(EB)与ct-DNA的光谱学作用.在ct-DNA存在时,berberine和EB的荧光强度都有较大的提高,pH的改变对二者荧光光谱的发射峰位与强度影响不大;紫外-可见吸收光谱随ct-DNA浓度的增加,表现出减色效应;二者都使ct-DNA的圆二色谱正负峰的吸收强度有所增加;琼脂糖凝胶电泳实验表明,EB-DNA体系的发光强度高于berberine-DNA体系.在DNA的定量测定中,可用无毒、副作用的小檗碱代替有致癌性的溴化乙锭,为安全、无环境污染、定量测定DNA提供了新试剂使用基础.The spectroscopy interactions between berberine and DNA, ethidium bromide and DNA are studied by UV-absorption, fluorescence, circular dichroism spectroscopy and agarose gel electrophoresis methods. When DNA exists, the fluorescence intensity of berberine and EB have a great increase, but the change of pH value has not great effect on the wavelength and intensity of the fluorescence. The absorbance of UV spectra present a hypochromic effect with DNA concentration increasing. The CD spectra of DNA has a great change both in negative and positive spectra. In the agarose gel electrophoresis, the luminescence intensity of EB-DNA system is more sensitive than berberine-DNA system. In the quantitative determination of DNA, the berberine rather than EB is implored, which can offer an important base of new reagent in the determination of DNA, and also afford clue to search more sensitive probe of DNA.
