ISOLATION AND IN VITRO STUDY OF HEPATIC SINUSOIDAL ENDOTHELIAL CELLS IN RAT

摘  要:应用胶原酶经门静脉插管灌注、Percoll密度梯度离心及体外培养等方法,从大鼠肝分离出高纯度的肝血窦内皮细胞,纯度达90%以上.电镜观察细胞表面有大量圆形窗孔,胞质内有丰富的穿内皮通道.肝血窦内皮细胞可与抗体包被的羊红细胞粘着形成花环,表明细胞表面保存良好的Fe受体.组织化学或免疫组织化学研究表明,培养24h的肝血窦内皮细胞里内皮素-1、一氧化氮合成酶、TGF-β阳性,而第Ⅷ因子相关抗原则为阴性.结果提示,肝血窦内皮细胞具有抗原呈递、参与肝微循环调节以及调控肝细胞增殖等功能.Using perfusion of collagenase via portal vein, isopycnic sedimentation in porcoll solution and tissue culture method, great number of hepatic sinusoidal endothelial cells (HEC) were isolated from rat livers. Under electron microscope, many fenestrae on cell surface (by SEM) and transendothelial channels in cytoplasma (by TEM) were clearly seen. In the co-culture of HEC with sheep erythrocytes coated by IgG, large numbers of rosettes were formed, indicating that HEC possessed the active Fc receptor on the surface. The results of histochemistry and immuno-cytochemistry assay showed that HEC exhibited endothelin (ET-1), NOS and TGF-β were positive, but FVIII related antigens were negative. These results suggest that the HEC expresses the important function of antigen and that its secretory products (ET, NO, TGF-β) may be involved in the regulation of the liver microcirculation and the proliferation of hepatocytes in physiological and pathological conditions of the liver.
