黑热病快速ELISA诊断方法的研究

摘　　要：用RPMI 1640纯培养的前鞭毛体可溶性抗原预固相PVC载体,用抗人γ链McAb制备酶结合物、TMB底物系统,建立了快速诊断黑热病的ELISA方法。该法与常规ELISA平行检测37例骨髓穿刺证实的黑热病患者血清,阳性符合率均为100%。检测120例健康人血清,均未见假阳性反应;与血吸虫、间日疟、囊虫、旋毛虫、华枝睾吸虫等寄生虫病人血清均未见交叉反应,但与麻风病人血清有10%的交叉反应,结果表明,该法具有较高的敏感性和较好的特异性。可与经典ELISA相比,但本法的操作更简便和快速。A kit consisted of PVC film prepared by coating antigen from cultured promastigotes in RPMI 1640, enzyme conjugate of anti-human gamma chain McAb and TMB substrate, was used for rapid diagnosis of leishmaniasis and the results were compared with those by routine ELFSA. Sera from 37 cases of leishmeniasis identified by bone marrow smear examination were tested with a result of 100% positive reaction rate. No false positive reaction was observed among 120 serum samples from normal individuals. Cross reactions were not found between leishmaniasis and schistosomiasis, vivax malaria, cysticercosis, trichinosis as well as clonolchiasis, except leprosy which had a 10% positive reaction rate among 30 cases. The results showed that the ELISA kit for rapid diagnosis of leishmaniasis has high sensitivity, good specificity and can be compared with the routine one but is much easier and quicker to perform.
