摘 要:目的研究异丙酚预先给药对脑缺血再灌注损伤大鼠脑组织葡萄糖转运体-3(GLUT3)表达的影响。方法健康雄性SD大鼠90只,体重290-310 g,随机分成3组:假手术组(S组,n=6)、生理盐水组(NS组,n=42)、异丙酚组(P组,n=42)。NS、P组采用线栓法阻断大脑中动脉1 h行再灌注,制备大鼠局灶性脑缺血再灌注模型,P组在缺血前10 min腹腔注射异丙酚10 mg/100 g,NS组给予等量生理盐水,S组只作切口,不行缺血。NS、P组分别在再灌注即刻、2 h、5 h、11 h、23 h、71 h、1周各处死6只大鼠,S组于实验11 h处死大鼠,断头取脑,计算脑梗塞体积比,用RT-PCR法测定GLUT3 mRNA表达水平,用免疫组织化学法测定GLUT3蛋白表达水平。结果NS组及P组再灌注期脑梗塞体积比、GLUT3 mRNA及GLUT3蛋白表达均高于S组,与NS组比较,P组脑梗塞体积比降低,脑组织GLUT3 mRNA及GLUT3蛋白表达升高(P<0.05)。结论异丙酚预先给药可上调脑缺血再灌注损伤大鼠脑组织GLUT3的表达,可能是其减轻大鼠脑缺血再灌注损伤的机制之一。Objective To investigate the effect of propofol pretreatment on the expression of glucose transporter-3(GLUT3) in the brain after cerebral ischemia-reperfusion (I/R). Methods Ninety male SD rats weighting 290-310 g were randomly divided into 3 groups: group Ⅰ sham operation( n = 6) ;group Ⅱ cerebral I/R (n = 42) and group Ⅲ propofol + cerebral I/R ( n = 42). The animals were anesthetized with intraperitoneal chloral hydrate 350μg·kg^-1 . A nylon thread ( diameter = 0.25 mm) with rounded tips was inserted into internal carotid artery and threaded cranially until resistance was felt. Mid-cerebral artery occlusion (MCAO)was confirmed by ipsi-lateral Homer's syndrome and contralateral heinplegia. After one hour MCAO the nylon thread was withdrawn to allow perfusion. In group Ⅰ the internal carotid artery was exposed but no MCAO was performed. In group Ⅲ propofol 10μg·100 g^- 1 was given intraperitoneally 10 min before MCAO while in group Ⅱ normal saline was given instead of propofol. The animals were killed at 0, 2, 5, 11, 23, 71 h and 1 week after reperfusion was started. Brains were immediately removed and sliced. The infarct size was estimated by using Kontron IBAS 2.5 image auto-analysis system. The GLUT3 mRNA was measured by RT-PCR. The expression of GLUT3 protein was determined by immuno-histochemistry. Results The infarct size was significantly smaller in group Ⅲ than in group Ⅱ . GLUT3 mRNA and GLUT3 protein started to increase at 2 h of reperfusion and remained significantly increased at lweek of reperfusion as compared to the borderline values immediately before reperfusion in both group Ⅱ and Ⅲ and were significantly higher in group Ⅲ than in group Ⅱ at all time points following reperfusion. Conclusion Propofol pretreatment can protect the brain from I/R injury to some extent. The up-regulation of GLUT3 expression is involved in the mechanism.
