摘 要:目的以同种异体神经移植修复大鼠坐骨神经缺损,探讨不同条件对同种异体移植神经及其支配骨骼肌病理变化的影响。方法以新鲜的和冷冻处理的同种异体神经移植修复大鼠10 mm坐骨神经缺损,术后实验组应用环孢素A治疗,8 w后行移植神经和其支配骨骼肌的病理学检查。结果新鲜的异体移植神经再生不良,骨骼肌萎缩明显,恢复最差;而应用环孢素A则最好;冷冻处理的神经移植中,应用免疫抑制剂的效果也优于未应用者;结论免疫抑制剂应用于新鲜的同种异体神经移植,在促进神经再生的同时也促进了骨骼肌的恢复。Objective: To study the effects of different conditions on nerve and skeletal muscle after sciatic: nerve allograft in rats. Methods: Thirty-two rats were equally divided into two groups, groups A and B, at random, in each animal, bilateral 10mm sciatic nerve defects was made, and left sciatic nerve was bridged with fresh nerve al lograft and the right side with nerve aliograft frozen in liquid nitrogen. The rats in the group B were giw:n cyclospofine A. Eight weeks after operation, the aliografted nerve and musculus tibialis anterior were pathologically examined. Results: Nerve regeneration and the skeletal muscle recovery in fresh aliograft without immunosupression was the poorest among these groups, but the best in fresh allograft under immunosupression with CSA. Nerve regeneration in frozen nerve allograft with CSA was better than that without CSA. Conclusion: Immunosupression is helpful to suppress the rejection of nerve allograft and improve the peripheral nerve regeneration and skeletal muscle recovery.
