摘  要:目的研究妊娠期高血压疾病患者腹腔巨噬细胞CD54和TNF-αmRNA的表达变化,探讨它们在妊娠期高血压疾病发病机理中的作用。方法体外培养30例妊娠期高血压疾病患者和10例正常孕妇腹腔液中的巨噬细胞,分别用流式细胞术半定量和逆转录多聚酶链反应(RT-PCR)方法测定实验组及对照组腹腔巨噬细胞表面CD54+及细胞内TNF-αmRNA的表达。结果轻度子痫前期、重度子痫前期组患者腹腔巨噬细胞CD54+细胞百分率较正常对照组明显增高,差异均有显著性(P<0.05);妊娠期高血压疾病组腹腔巨噬细胞TNF-αmRNA表达水平均明显高于对照组(P<0.05);相关性分析显示,CD54表达量与TNF-αmRNA含量呈正相关。结论在妊娠期高血压疾病中,巨噬细胞高表达CD54与TNF-αmRNA,二者表达量呈正相关;CD54与TNF-α可能是参与妊娠期高血压疾病的重要因子。Objective:To study the expression of CD54 and tumor necrosis factor-alpha (TNF-α) mRNA in peritoneal macrophages of hypertension disorder complicating pregnancy (HDCP) and the potential role of these cytokines. Methods:Peritoneal macrophages were obtained from 30 patients with HDCP and 10 normal pregnants. CD54 was measured by flow cytometry. Reverse transcription-polymerase chain reaction(RT-PCR) was used to detect TNF-α mRNA. Results: The percentage of CD54 positive cells in mild preclampsia and severe preclampsia groups was significantly higher than that in control ( P 〈 0.05 ). The levels of TNF-α mRNA in peritoneal macrophages of HDCP group were significantly higher than that in control group(P 〈0.05). There was significantly positive correlations between the expression of CD54 and TNF-α mRNA in peritoneal macrophages. Conclusion:The expression of CD54 and TNF-α mRNA in peritoneal macrophages of hypertensive patients were up-regulated, CD54 and TNF-α may be important factors in the pathogenesis of HDCP.
