环孢素对哮喘大鼠体内相关因子的影响

摘　　要：目的探讨环孢素对哮喘大鼠C-反应蛋白(CRP)、转化生长因子-β1(TGF-β1)、基质金属蛋白酶类(MMPs)及其抑制剂(TIMPs)表达的影响。方法将80只哮喘模型SD大鼠随机分为5组:正常对照组,哮喘组,地塞米松组,环孢素大、小剂量组。大鼠给予卵蛋白(OVA)致敏和刺激后成为哮喘模型,用环孢素大剂量10mg·kg-1·d-1,小剂量5mg·kg-1·d-1,地塞米松5mg·kg-1·d-1进行干预,采血检测及肺组织标本经免疫组化染色后计算机图像分析测定上述因子含量。结果哮喘组CRP、TGF-β1含量为(82.30±30.27)、(63.55±10.73)mg·L-1,环孢素小剂量组为(47.16±18.67)、(32.18±7.16)mg·L-1,正常对照组含量为(3.42±1.63)、(14.22±4.08)mg·L-1,哮喘组CRP、TGF-β1含量明显高于对照组(P<0.05);MMP-9阳性率在哮喘组表达75.0%,环孢素小剂量组表达68.8%,明显高于正常组25.0%(P<0.05);TIMPs含量则明显低于正常对照组(P<0.05)。结论哮喘组CRP、TGF-β1、MMPs含量较正常对照组明显增高,TIMPs明显降低。大剂量组环孢素与激素组均可以使哮喘组中的CRP、TGF-β1、MMPs及TIMPs表达趋于正常组,环孢素小剂量组对上述因子效果不明显。A/M To explore the significance of the level change of C-reactive protein （CRP）, transformin growth factor-beta one （TGF-β1）rnatrix metalloproteinase （MMPs） and tissue inhibitor of metalloproteases （TIMPs） in asthmatic rats before and after eielosporin＇ s apply. METHODS Eighty SD rats were divided into normal group, model group, desamethasone group, low-dose group and high-dose of ciclosporin group. The changes of CRP, TGF-β1, MMPs and TIMPs in samples were measured by the computerized image analysis system. RESULTS The contents of CRP, TGF-β1 in model groups （82.30± 30.27）mg·L^-1, （63.55 ± 10.73）mg·L^-1and in low-dose groups （47.16± 18.67）mg·L^-1 ,（32.18 ± 7.16） mg·L^-1, were significandy higher than in control groups （ 3.42 ± 1.63） mg·L^-1, （ 14.22 ± 4.08） mg·L^-1 （ p 〈 0.05） .The masculine rates of MMP-9 in model group and in low-dose of cyclosporin group were higher than in control group but the masculine rates of TIMPs were lower than in control group. CONCLUSION The contents of CRP, TGF-β1, MMPs in model group were higer than those in control group. High-dose of ciclosporin and desamethasone could make the expression of CRP,TGF-β1, MMPs and TIMPs in model rats go to normal.
