分泌型白细胞蛋白酶抑制剂对内毒素致新生大鼠急性肺损伤的抗感染作用

摘　　要：目的探讨分泌型白细胞蛋白酶抑制剂(SLPI)对内毒素(LPS)致新生大鼠急性肺损伤(ALI)的抗感染作用。方法利用LPS气管内滴入制备新生大鼠ALI模型,设立SLPI治疗组、LPS组、生理盐水(NS)对照组。SLPI治疗组在气管内滴入LPS前30 min预先滴入400μg SLPI。4 h后取各组大鼠肺组织匀浆,用琼脂扩散法、ELISA法分别测定中性粒细胞弹性蛋白酶(NE)活性和TNFα-水平。结果LPS组肺组织NE活性、TNFα-及总蛋白水平均显著高于SLPI治疗组和对照组,3组比较差异有显著性(F=1475.28,35.8,21.6 P均<0.01);SLPI治疗组肺组织NE活性、TNF-α及总蛋白水平均显著低于LPS组,差异有显著性(q=66.14,7.02,5.60 P均<0.01);但高于对照组,亦有显著性差异(q=22.18,5.07,3.77 P<0.01,0.05)。结论SLPI对LPS致新生大鼠急性肺损伤有抗感染作用。Objective To investigate the anti - inflammatory effects of secretory leukocyte protease inhibitor（SLPI） on newborn rats with lipopolysaccharide（LPS） - induced acute lung injury（ALl）. Methods Newborn rats model of ALl induced by intratracheal instillation of with a dose of l0 mg/kg. Rats were randomly divided into SLPI treatment group, LPS group and NS control group. SLPI treatment group received an intratracheal administration of 400 pg SLPI at 30 min before they got LPS. At 4 hours after they received SLPI, pulmonary tissues were obtained and homogenated. Agar diffusion method and ELISA were used to detecte neutrophil elastase（NE） activity and concentration of tumor necrosis factor - α（TNF - α）, and total proteins respectively. Results The significant differences were found in NE activity, TNF a and total protein measurement among 3 groups（ F = 1475.28,35.8,21.6 all P 〈 0.01 ） ; NE activities, TNF-α levels and total proteins were significantly higher in LPS group than those in SLPI treatment group and normal control group. NE activities and TNF - α levels and total proteins were remarkably lower in SLPI treatment group than those in LPS group, but higher than those in normal control group（ P 〈 0.01 ）. Conclusion SLPI can inhibit LPS - induced inflammation in lung of newborn rats.
