摘  要:目的:高效液相色谱法测定大鼠血浆中头孢氨苄浓度。方法:血浆样品用超滤法去除蛋白质直接进样。以μBondapak^TM C18柱(4.6mm×250mm,5μm)为色谱柱,水-甲醇(400:100,v/v,用1.0mol/L醋酸调pH至4.0)为流动相,流速为0.8ml/min,柱温25℃;二极管阵列检测波长为254nm。结果:头孢氨苄浓度在0.63~10.0μg/ml范同内线性关系良好,相关系数〉0.9999;其日内和日间相对标准差(RSD)〈10%(n=5),方法回收率为98.1%~100.1%(n=5)。结论:该法快速简便,适用于临床血药浓度研究和间接评价结肠上皮二肽载体(PepT1)的功能。Objective : A high performance liquid chromatographic method for the determination of Cefalexin in rat plasma was developed. Methods: The plasma sample was deproteinized by an uhrafihration. A reversal phase ( μBondapak^TM 4.6 mm ×250 mm, 5 μm) column was used with a mobile phase of water-methanol (400 : 100, v/v ; pH 4.0) and flow rate of 0.8 ml/min. The photodiode array detector was used. Results: A good linearity was obtained from 0.63 - 10.0 μg/ml of Cefalexin, the relation coefficient was more than 0. 999 9. The relative standard derivations for within-day and between-day were all 〈 10% ( n = 5 ), and the recoveries were 98.1% - 100.1% ( n = 5 ). The average recoveries of method ( n = 5 ) was 98.1%. Conclusion : This method was simple and rapid, and was suited for clinical investigation of Cefalexin in rat plasma.
