摘 要:探讨用灵长类动物树鼩建立呼吸道合胞病毒(RSV)感染动物模型的可行性。28只树鼩随机分为7组,每组4只,鼻内滴入10^6PFU RSV,感染后每24h检测1组:另取7只树购鼻内滴入100μl Hep-2细胞培养液,滴鼻后每24h检测1只作对照:无菌取树鼩肺组织进行病毒分离、空斑形成实验检测病毒滴度、病理检查和RT-PCR检测肺组织内RSV mRNA表达:结果显示:树鼩感染RSV后,无明显呼吸道感染症状;树鼩肺组织匀浆接种于Hep-2细胞上,第3、4、5实验组出现细胞病变效应(CPE);树鼩感染RSV后肺部病理改变在3~7d较为明显,主要为间质改变;在10^6 PFU感染浓度下树鼩肺组织内RSV处于低水平复制,复制的高峰期在3~5d,峰值在第4d。提示:树鼩可以用来建立RSV感染的动物模型。To establish a novel model for respiratory syncytial virus(RSV) infection in primate animal, twentyeight tree shrews were divided into seven groups and inoculated through intranasal route with 100μl RSV of 10^7 plaque forming units (PFU). Another seven tree shrews set up as control received 100μl Hep-2 cell culture fluid. At every 24h tree shrews in one experimental group and one in control group were killed. Their right lungs were removed under aseptic condition, homogenized and centrifuged, the supernatants were collected for virus isolation and quantification of virus consecutively. Left lungs were collected for histopathological examination and detection of expression of RSV mRNA by RT-PCR. Results showed that: (1)Tree shrews infected by RSV did not show any respiratory symptom; (2)The lung homogenate of tree shrews when inoculated onto Hep-2 cell, the cytopathogenic effect was observed in 3th, 4th, 5th experimental groups; (3)The pathological changes were obvious at day 3 to day 7, the most apparent change was interstitial pneumonitis; (4)Inoculated with 10^6PFU RSV, viral replication in tree shrew lungs were at low level, replication reached to maximum within 3 to 5 days, peak at day 4. The results suggest that tree shrew have potential for establishing RSV infection animal model.
