摘　　要：目的以本实验室研制的重组人自身抗原IA-2作为抗原,建立血清中抗IA-2的自身抗体的ELISA测定方法,并对正常人群进行检测。方法在间接ELISA法建立过程中对封闭剂、血清稀释度及二抗稀释度及其他测试条件进行了选择,方法建立之后,对110名正常人血清中的IgG型抗IA-2抗体进行了检测,用国外已成型的试剂盒进行了对照性实验,并进行统计学分析。结果确立了血清中抗IA-2自身抗体的间接ELISA法,以本方法对110例正常人血清中抗IA-2自身抗体进行检测的平均滴度为0·31±20·07,与用进口试剂盒进行检测的结果进行配对t检验,无显著性差异。结论建立了血清中抗IA-2自身抗体的检测方法及正常值为糖尿病的初筛、早期诊断的开展,做了准备。Objective To establish a quantitative ELISA for detection of anti-IA-2 antibody in using the recombinant human IA-2 as antigen and to define the normal vaule of anti-IA-2 antibody in our laboratory. Methods After a series of preliminary tests, the proper covering substance,sarape diluting degree and concentration of enzyme-linked goat anti human IgG were screened and selected. And then, the IA-2 antibody of 110 healthy people was examined in using the above method. Results A quantitative ELISA for detection of anti-IA-2 antibody in sera of persons was established and the normal value of the average OD450 （x±s） was 0. 31±2 ＊0. 07. Conclusions A quantitative ELISA and its normal value for detection of anti IA-2 antibody in diabetes patients are established.
