摘  要:目的:分析中药复方温胆汤改善睡眠的作用与脑肠肽-胆囊收缩素8表达的关系。方法:实验于2004-09/2005-01在黑龙江中医药大学中医方药分析实验室完成。选择健康雄性Wistar大鼠28只,按随机数字表法分为4组,即正常对照组、对氯苯丙氨酸化失眠模型组、温胆汤组及地西泮组,每组7只。除正常对照组外,其余3组大鼠腹腔注射对氯苯丙氨酸复制大鼠失眠模型,动物出现昼夜节律消失,白天也活动不停,整体观察与正常对照组有明显不同,表明模型复制成功。模型复制成功后,每日8:00am按1mL/kg体积灌胃给药,温胆汤组大鼠给予600g/L温胆汤水煎液(由半夏10g,竹茹10g,枳实10g,橘皮15g,甘草5g,茯苓7.5g,生姜5片、大枣1枚(5g)组成,6.1g/kg生药);地西泮组给予92g/L地西泮水溶液;正常对照组和对氯苯丙氨酸化失眠模型组给予蒸馏水,1次/d,共6d。于末次给药50min后,将大鼠直接脱颈椎处死,取所需脑组织,按照免疫组织化学方法处理后,在CMIAS多功能真彩色病理图像分析系统下分析温胆汤对失眠大鼠皮质及下丘脑胆囊收缩素8表达的影响。结果:28只大鼠全部进入结果分析,无脱失。各组大鼠大脑皮质及下丘脑胆囊收缩素8阳性细胞百分率比较:对氯苯丙氨酸化失眠模型组大鼠皮质、下丘脑胆囊收缩素8阳性细胞百分率均显著低于正常对照组[(3.35±0.33)%,(15.14±0.69)%;(5.95±0.84)%,(27.96±2.42)%,t=3.6003,12.8119,P<0.01],温胆汤组大鼠皮质、下丘脑胆囊收缩素8阳性细胞百分率均显著高于对氯苯丙氨酸化失眠模型组[(5.47±0.89)%,(24.60±1.77)%,t=2.1211,9.4600,P<0.01]。地西泮组大鼠皮质、下丘脑胆囊收缩素8阳性细胞百分率与对氯苯丙氨酸化失眠模型组相近[(4.17±0.18)%,(16.82±1.70)%,P>0.05]。结论:中药复方温胆汤可以明显增强失眠大鼠大脑皮质、下丘脑胆囊收缩素8的阳性表达,进而增加大鼠睡眠,推测胆囊收缩素8可能是“胃不�AIM: To analyze the relation between the ameliorating effect of traditional Chinese compound Wendan Tang on sleep and the expression of braingut petide-cholecystokinin (CCK8). METHODS: The experiment was conducted at the Analytical Laboratory of Chinese prescription, Heilongjiang University of Traditional Chinese Medicine from September 2004 to January 2005. Twenty-eight healthy male Wistar rats were selected and randomly divided into four groups: normal control group, para-chlorophenylalanine insomnia model group (PCPA group), Wendan Tang group (W group) and diazepam group (D group) with 7 rats in each group. All rats except those in the normal control group were intraperitoneally injected with PCPA to made into rat models of insomnia. After that symptoms represented in rats as the following: circadian rhythm disordered with activities at night. General observation showed significant differences in comparison with the normal control group, which suggested successful modeling. Successful rat models were gastrieally perfused of lmL/kg at 8:00 Am daily, and rats in the W group were given 600 g/L Wemlan Tang (composed of 10 g of pinelliae tuber, 10 g of eaulis bambusae in taeniam, 10 g of aurantii fruetus immaturus, 15 g of tangerine pericarp, 5 g of licorice root, 7.5 g of hoelen, 5 pieces of ginger and 5 g of fruetus jujubae, 6.1 g/kg crude drug). Rats in the diazepam group were given 92 g/L diazepam water solution. Rats in the normal control group and PCPA group were given distiIled water, once a day for 6 continuous days. Rats were killed at 50 minutes after the last administration directly by shedding off the cervical vertebrae, and the brain tissues were obtained and handled with immunohistochemieal method. The effects of Wendan tang on the expression of CCK8 in the cortex and hypothalamus of insomnia rats were analyzed by CMIAS multifunetional coJor image analytical system. RESULTS: A total of 28 rats were involved in the analysis of results, and no one withdrew from the study.
