摘 要:目的:观察以单味使用具有较弱的收敛、敛疮、抗菌、消炎等多种功能的中药组方的复方儿茶水提液抗腹泻和抗炎作用,寻找高效低毒的抗腹泻药。方法:实验于2005-07/12在大连大学药理实验室完成,选择清洁级昆明种小鼠280只,体质量18~22g,雌雄不拘。随机数字表法分为复方儿茶水提液2,4g/kg剂量组、阳性药对照组、生理盐水组4组,每组70只。分别灌胃复方儿茶水提液2,4g/kg,阳性药(黄连素0.03g/kg,硫酸阿托品10mg/kg,阿司匹林0.3g/kg,以等容积生理盐水制成药液),同容积生理盐水。每组再均分为7个亚组,分别施加番泻叶(2g/kg)、蓖麻油(7.5g/kg)、大黄(20g/kg)、硫酸钠(20g/kg)和二甲苯(0.05mL/只)、乙酸(0.7g/kg)7个因素致小鼠腹泻或炎症模型,观察复方儿茶水提液的抗腹泻和抗炎作用。结果:纳入280只小鼠,均进入结果分析。①复方儿茶水提液2,4g/kg剂量组给药后1,2,3,4h番泻叶、蓖麻油、大黄、硫酸钠所致腹泻次数显著低于生理盐水组(P<0.05~0.001),复方儿茶水提液4g/kg剂量组作用优于2g/kg剂量组。②复方儿茶水提液2,4g/kg组胃肠推进率与生理盐水组比较差异有非常显著性意义(P<0.01~0.001),与阳性药(硫酸阿托品10mg/kg)对照组比较差异没有显著性意义(P>0.05)。复方儿茶水提液2,4g/kg组、阳性药对照组抑制率分别为40.0%、78.6%和34.4%。③复方儿茶水提液2,4g/kg组、阳性药(阿司匹林)对照组左右耳片质量差均显著低于生理盐水组(P<0.05~0.01)。④复方儿茶水提液2,4g/kg组和阿司匹林组对乙酸提高的小鼠腹腔毛细血管通透性抑制率分别为42.5%,64.9%和66.7%(P<0.05~0.01)。结论:复方儿茶水提液抗腹泻作用机制广泛,具有抗渗透性泻下和刺激性泻下作用,可以通过抗炎对抗炎症介质的泻下作用,产生抗腹泻作用。这些抗炎作用可能是复方儿茶水提液抗腹泻作用的重要机制。AIM:To observe tbe antidiarrheal and anti-inflammatory effects of water extraction solution of catechu composita (WESCC), which has multiple functions of weak stypsis, ulcer-control, antibiosis and eliminating inflammation, and look for antidiarrheal drugs with high effects and low toxicity, METHODS:The experiment was carried out in the Department of Pharmacology, Medical College of Dalian University from July to December 2005, and 280 female and male Kunming mice of clean grade weighed 18-22 g were selected and randomly divided into 2, 4 g/kg WESCC group, positive control group and normal saline group with 70 mice in each group. Mice received gastric perfusion of 2, 4 g/kg WESCC, positive drugs (0.03 g/kg berberine, 10 mg/kg atropine sulfate and 0.3 g/kg aspirin were made into physic liquor by dissolving in samevolume of normal saline) and same-volume of normal saline respectively. Each group was subdivided into 7 subgroups. Senna (2 g/kg), castor oi1(7.5 g/kg), rhubarb(20 g/kg), sodium sulfate(20 g/kg), xylene (0.05 mL/mouse) and acetic aeid (0.7 g/kg) were used to induce mouse models of diarrhea or inflammation to observe the antidiarrheal and antiinflammatory effects of WESCC. RESULTS:All the 280 enrolled mice were involved in the analysis of results. ①The diarrheal frequency of mice in the 2 g/kg and 4 g/kg WESCC group induced by senna, castor oil, rhubarb and sodium sulfate at one, two, three, four hours after administration was significantly lower than the normal saline group (P 〈 0.05-0.01), and the effects of 4 g/kg WESCC group was better than 2 g/kg WESCC group. ②There were remarkable differences in gastro intestinal propellant rate between 2, 4 g/kg WESCC groups and normal saline group (P 〈 0.01-0.001), while there was no significant differences in comparison with positive control group (10 mg/kg atropine sulfate) (P 〉 0.05). The inhibition ratio in 2,4 g/kg WESCC group and positive control group was respectively 40.0%, 78.6% and 34.4%.③
