摘  要:目的:观察脾阴虚证衰老模型大鼠心肌、脑组织线粒体DNA缺失及线粒体呼吸链酶复合体活力的变化。方法:实验于2002-07/2004-05在辽宁中医学院实验动物中心完成。①选用健康3月龄SD大白鼠20只为青年对照组,雌雄各半。选用40只21月龄SD大白鼠(雌雄各半),将其按随机抽签法分为2组:老年对照组和模型组,每组20只。②模型组先对老年大鼠施以饮食不节加劳倦过度造成脾气虚证模型,然后在其基础上,每日增加伤阴药(附子、吴茱萸,按1∶1比例,购于辽宁中医学院附属医院)10mL/kg胃饲。老年对照组和青年对照组予同体积的生理盐水灌胃。③动物造模实验第90天,用TanonGIS凝胶图像处理系统对全部PCR扩增产物电泳后条带进行吸光度(A值)扫描,检测心肌、脑组织线粒体DNA缺失产物A值。参照伍期专等的方法测定心肌、脑组织线粒体呼吸链酶复合体活力。④两组间均数比较采用t检验;多组间均数比较采用单因素方差分析。结果:脾阴虚证造模时,死亡11只,模型评估后,为了便于统计,最终纳入分析为每组7只,共21只。①大鼠心肌、脑组织线粒体呼吸链酶复合体活力:老年对照组和模型组明显低于青年对照组(P<0.05~0.01),模型组明显低于老年对照组(P<0.05~0.01)。②大鼠心肌、脑组织线粒体DNA缺失产物A值:老年对照组和模型组明显高于青年对照组(心肌:81.29±16.65,178.71±9.98,0.14±0.38鸦脑组织:90.85±6.82,163.29±11.63,1.85±4.49,P<0.01),模型组明显高于老年对照组(P<0.01)。结论:脾阴虚衰老机体细胞线粒体DNA缺失明显增多,线粒体呼吸链酶复合体活力明显下降。AIM: To observe the changes of activities of mitochondrial respiratory chain complexes and the deficiency of mitochondrial DNA in myocardium and brain tissues of aged rats with spleen yin deficiency syndrome. METHODS: The experiment was conducted in the Experimental Center of Animals, Liaoning College of Traditional Chinese Medicine between July 2002 and May 2004.①Twenty healthy SD rats of 3 months old (half males and half females) were selected as the young control group. Forty SD rats of 21 months (half males and half females) were selected and randomly divided into 2 groups: old control group and model group with 20 rats in each group. ②Rats model of spleen yin deficiency syndromes were established in the model group by uncontrolled diet and overstrain, based on which 10 mL/kg of damaged yin drugs (aconite root and evodiae, fructus at the ratio of 1:1, brought from the Affiliated Hospital of Liaoning College of Traditional Chinese Medicine) were administrated to rats. Rats in the old control group and young control group were intragastrically administrated with normal saline at the same volume. ③On the 90^th day of modeling, Tanon Gis gel image processing system was adopted to conduct absorbance scanning (A value) on the straps of all PCR amplification products after electrophoresis, and the A value of deficiency of mitochondrial DNA in myocardium and brain tissues were detected. The activity of mitochondrial respiratory chain complexes in myocardium and brain tissues were determined by methods of Wu etc.④Differences in mean between two groups were compared with t test, and differences among groups were compared with single-factor analysis of variance. RESULTS: Eleven rats died in the modeling.
