摘  要:目的筛选与精子发生相关的基因。方法将不同发育阶段的小鼠睾丸组织cDNA探针与Affymetrix全基因组芯片进行杂交,通过杂交信号的比较,筛选出差异表达的基因。利用网络信息资源对该基因进行生物学信息分析。RT-PCR分析该基因在小鼠不同发育阶段睾丸及小鼠不同组织中的表达。结果通过4、91、8、35、54、日龄和6月龄小鼠睾丸的芯片信号比较筛选出一个差异表达杂交点(GenBank登录号:NM-026234),生物信息学分析发现该基因全长3 979 bp,含有1 272 bp的完整ORF,编码一个有423个氨基酸、分子量为49.788的蛋白质。PigM蛋白与大鼠同源基因PigM蛋白有96%的同源性,与人PigM蛋白有89%的同源性,显示该基因在哺乳动物中高度保守。RT-PCR分析表明PigM基因特异性表达于睾丸组织及18日龄后的小鼠睾丸中。结论PigM基因的表达与小鼠精子发生的过程一致,显示其可能在精子发生中起着重要作用。解释该基因的生物学功能,可对揭示人类无精、弱精和少精引起的男性不育的病因,治疗和预防男不育及寻找新的男子避孕药的靶位点具有重要意义。Objective To identify genes involved in spermatogenesis. Methods Testes eDNA samples from Balb/C mice of different postnatal days were performed with mouse whole genome Affymetrix chip analysis to screen the testis-specific genes. The expression profile of the selected gene was also identified by RT-PCR. The characteristics of the selected gene were analyzed by various bioinformatie tools. Results By analysing the hybridization signals, a gene PigM with a differential expression in the developmental stages of testis was identified. The full length eDNA of 3979 bp contained an open reading frame of 1272 bp which encoded a putative , protein of 423 amino acids. RT-PCR analysis revealed that PigM mRNA was exclusively expressed in mice testis. The protein of the homologous gene PigM in human has 89% identity to that of the PigM in mice while the protein of PigM in rat has 96% identity. Conclusion The present study suggests that PigM functions may be as a testis-specific transcription factor that plays important roles during spermatogenesis. Well understanding of the molecular function of Pig, M, which is often based on the treatment of male sterility due to abnormity of sperm development, and offering new contraceptive targets and health care drugs.
