当归、阿魏酸钠对骨关节炎软骨退变与自由基的影响  

Effects of Angelica and Sodium Ferulate Injection on Cartilage Degeneration and Metabolism of Free Radicals in Rats with Osteoarthritis

摘　　要：目的:探讨当归、阿魏酸钠关节腔注射对大鼠骨性关节炎(OA)软骨退变与自由基的影响。方法:4%木瓜蛋白酶关节腔注射建立大鼠OA模型,1周后48只大鼠分成6组:正常组,模型组,25%、5%当归治疗组,0.5%、0.1%阿魏酸钠治疗组,分组后即开始用不同浓度的当归、阿魏酸钠关节腔注射治疗,每3 d 1次,每次双膝各注射0.1 ml,对照组注射生理盐水。6周后处死动物,抽取血液、关节液检测SOD、MDA,切取关节,大体及组织学观察。结果:两药在高浓度可明显修复关节软骨的退变(均P<0.01),降低关节液、血浆的MDA水平(均P<0.01),增加SOD活性(均P<0.05);两药在低浓度时SOD升高不明显(均P>0.05),但阿魏酸钠仍可明显修复关节软骨的退变(均P<0.01),降低MDA的水平(P<0.01,P<0.05)。结论:当归、阿魏酸钠关节腔注射治疗大鼠OA可促进退变软骨的修复,阿魏酸是当归注射液治疗OA的有效的单体成份之一。Objective： To explore the effects of angelica injection and sodium ferulate（SF） injection on cartilage degeneration and free radicals in rats with osteoarthritis. Methods： Animal models of osteoarthritis were made in rats by intraarticular injection of 4 % papain. Forty-eight rats were divided into six groups （eight in every group） ： normal group, model group, 25% and 5% angelica injection treatment groups, 0.5% and 0.1% SF injection treatment groups. The animals began receiving angelica injection and SF injection of different concentration every three days by intraarticular injecting one week after the last injection of papain and were killed six weeks later. Macroscopic and histological analysis were performed on the articular cartilage. In the same time, the contents of MDA and the activity of SOD in the plasma and synovial fluid were detected. Results： Both drugs of high concentration could significantly repair cartilage degeneration （both P〈0.01） and reduce the levels of MDA （both P〈0.01）and enhance the SOD activity（both P〈0.05）. Both drugs of low concentration had no significant effect on SOD activity, but 0.1% SF could repair cartilage degeneration （P〈0.01） and reduce the level of MDA in plasma and in synovia （P〈0.01 and P〈0.05 respectively）. Conclusion： Both angelica and sodium ferulate injection have beneficial effect on repairment of the involved articular cartilage degeneration of the knee with experimental osteoarthritis in rats. Ferulate is one of effective components of angelica injection in treating osteoarthritis.
