摘 要:目的:探讨大鼠脊髓背根神经节(DRG)内是否存在神经前体细胞.方法:取成年和胚胎大鼠DRG冰冻切片,进行B rdu,Nestin,P75免疫荧光组织化学检测,分别取成年和胚胎大鼠DRG进行原代细胞培养,观察其增殖与分化情况,并行Nestin免疫组织化学和B rdu,P75,NF,GFAP免疫荧光组织化学鉴定.结果:在成年和胚胎大鼠DRG冰冻切片上均见有B rdu/Nestin和Nestin/P75免疫荧光双标细胞.细胞培养观察到取自DRG的细胞有分裂增殖,呈Nestin免疫组织化学阳性反应和B rdu,P75免疫荧光组织化学阳性反应;培养第三代细胞加入胎牛血清后出现分化,分化细胞呈NF和GFAP免疫荧光组织化学阳性反应.结论:大鼠DRG内存在有分裂增殖的神经前体细胞,这些细胞可以分化为神经元和神经胶质细胞.AIM: To investigate neural precursor cells in ra dorsal root ganglia (DRG). METHODS: DRG from embryonic and adult rats were frozen and sliced for Brdu, Nestin, p^75 immunofluoreseenee histoehemieal test. Additionally, the cells of ganglia were respectively cultured and observed in thei proliferation and differentiation. The cells were identified by Brdu, Nestin, p^75, NF and GFAP immunofluoreseenee histo. chemical test, RESULTS: In the frozen DRG sections from adult and embryonic rats, Brdu/Nestin and Nestin/P^75 positive eelh were found. The cultured cells could divide and proliferate am showed Nestin, Brdu, p^75 positive, The third-passage cells could differentiate in the presence of fetal bovine serum, The differentiating cells showed NF and GFAP positive. CONCLUSION: There are neural precursor cells that can divide and proliferate in rat DRG. They can differentiate into neuron and glial cells.
