摘 要:目的:观察自体微小颗粒骨复合骨髓基质细胞(MSC)或成骨细胞移植修复大鼠骨缺损的治疗效果.方法:制作大鼠颅骨缺损动物模型,培养同种异体新生大鼠的MSC及成骨细胞,复合自体颗粒骨后植入骨缺损区,X线摄片观察骨愈合情况;RT-PCR检测转化生长因子β1(TGF-β1),碱性成纤维细胞生长因子(bFGF)表达水平.结果:植入微小颗粒骨复合成骨细胞组颅骨缺损愈合最快(7.33±0.44)wk,TGF-β1,bFGF表达出现早,与其他两组比较差异显著(P<0.05).植入微小颗粒骨复合MSC组颅骨缺损愈合时间居中(8.67±0.69)wk,TGF-β1,bFGF表达早于单纯微小颗粒骨组(P<0.05).单纯微小颗粒骨植入组颅骨缺损愈合慢(11.67±1.52)wk,TGF-β1,bFGF表达出现晚.结论:微小颗粒骨复合细胞移植修复颅骨缺损,细胞因子表达早,缺损愈合明显加快.AIM: To investigate the combined treatment effects of autogenetic minimal morselized bone and allogenic cells on rat cranial defect. METHODS: Cranial defect in rat models was repaired with autogenetic minimal morselized bone graft and allogenic cell transplantation [ osteoblasts or marrow stormal cells (MSC) ]. X ray was taken to understand the union condition, and RT-PCR was performed to examine the mRNA expressions of TGF-β1 and bFGF. RESULTS: In the order of combined treatment group A (morselized bone and osteoblast graft), combined treatment group B (morselized bone and MSC graft ) , combined treatment group C( minimal morselized bone only) , the union time of cranial defect prolonged as (7.33±0.44) wk, ( 8.67 ± 0.69 ) wk and (11.67 ± 1.52) wk, and the expression of TGF-β1 and bFGF emerged in the order ( P 〈 0.05 between every 2 groups ). CONCLUSION: The combined therapy of minimal morselized bone and allogenic cells accelerate the repairment of cranial defect and the expression of cell factors.
